- Executive summary:
The boiling point of the test item was determined with the distillation method according to recognised guideline and GLP. No deviation from the guideline was observed during the study.
A preliminary test was performed to determine the experimental conditions to be used in the main tests. Based on the results obtained in the main tests, the boiling range of the test item is 70.3°C and 72.8°C at reduced pressure (0.13 Pa). While boiling, the test item is partially decomposed.
In the preliminary test, in a micro-distillation apparatus, a small amount of the test substance in a bottle was heated. The test substance was heated to 199°C at an atmospheric pressure of 101.89 kPa at which the test substance started to boil. The test substance started to distil at a distillation temperature of 83.5°C. The test substance distilled dropwise while the distillation temperature raised to 207.5°C. The distillation stopped and the test substance (residue) decomposed to a black/brown substance. Because the test substance decomposed above 60°C, the boiling temperature was determined under reduced pressure.
In the two main tests, a micro-distillation apparatus was used. The test substance was heated to 135 or 125°C under reduced pressure (0.13Pa) at which the substance started to boil. At a distillation temperature 70°C or 70.5°C the test substance started to distil dropwise. The test substance was distilled as a whole at a distillation temperature of 73°C or 72.5°C.
In order to determine if the test substance was decomposed during the test, the distillate was analysed by mean of HPLC. For this purpose, a certain amount of the distillate obtained in the main tests was dissolved in methanol. The solutions were analysed by means of HPLC. The analytical method used was calibrated for quantification of the test item between 1.3 and 212.9 mg/l. The recovery percents of the distillate obtained in the main tests were 72% and 56%. These results proved that partial decomposition of the test substance occurred during the test.
