Activated sludge was obtained from a sewage plant treating predominantly domestic sewage (Nanjing Chengdong Sewage Treatment Plant) (Batch No.: AS20I 707I2I).
The sludge was washed with basal mineral salt medium (BSM) by settling the sludge in a graduated cylinder, decanting the supernatant liquid, replacing with fresh BSM, aerating, and settling. The above washing process was repeated 3 times and was aerated until use.

Details on results:
The initial concentration of the test substance in the reaction vessels was 31 .6 mg/L and 30.0 mg/L. The microbial inoculum concentration (mixed liquor suspended solids) was 30 mg/L.
During the test, the temperature was kept at (22±1) °C. The pH value of the content in the test container was maintained between 6.28 and 7.38. The total oxygen uptake in the inoculum blank was 26.1 mg 02/L at the end of the study, ranged 20 ~30 mg O2/L.
Biodegradation of the reference substance, aniline, reached 80.3 % at 14 days (> 60%). The difference between replicate values was less than 20% during the study. Biodegradation of inhibition control was 54.4 % at 14 days(> 25%), and the oxygen consumption by the test substance is greater than 60% of inoculum blank during the test period, indicating that there was no inhibition effect to inoculum. Thus, the test was considered valid.

Executive summary:

The ready biodegradability of Methanaminium, N,N,N-trimethyl-, salt with 2,2-dimethylpropanoic acid (1: 1) was determined in a 28-day oxygen consumption test (OECD 301F) using activated sludge from a domestic waste water treatment plant as the source ofthe microbial inoculum.

Activated sludge, surface soil and surface water were sampled from ten sites distributed in four districts throughout Nanjing city, such as Chengdong, Chengbei, Jiangxinzhou and Jiangning. 1L of the sludge, soil and water were collected and mixed thoroughly together.
After removing floating matter, the mixture was allowed to stand and then the supernatant was filtrated through filter paper. After that the filtrate was adjusted to pH 7.0 with sodium hydroxide or phosphoric acid. Finally an appropriate volume of the filtrate was transferred to a fill-and-draw activated sludge vessel and aerated for about 23.5 h (Batch No.: IN201801041).

Test solutions were prepared in an inorganic salts medium, inoculated with a number of microorganisms collected from 10 places in Nanjing city. During the test, the temperature was kept at (25±2) °C. The test was valid because the level of biodegradation of the reference substance

aniline was 56.6 % after 7 days(> 40%), and 77.3 % after 14 days(> 65%), and the recovery rate of residual amount of the test compound in the "abiotic control" is found to be more than 10% after 28 days.

The BOD results showed that inherent biodegradation of the test substance (Methanaminium, N,N,N-trimethyl-, salt with 2,2-dimethylpropanoic acid (1: 1)) was 92.2% after 28 days.

Based on the residue analysis, at the end of the experiment, the inherent biodegradation rate was 94.8%.
