Experimental studies investigating the short-term toxicity for Amines, C12-14-tert-alkyl, mixed sec-Bu and iso-Bu phosphates (CAS No. 96690-34-5) are available for fish and aquatic invertebrates. The studies were performed under GLP conditions and according to standard OECD guidelines. For fish an LC50 value was determined to be at 18 mg/L (nominal loading rate, WAF) and for aquatic invertebrates the EL50 (48 h) was 1.13 mg/L (nominal loading rate, WAF).
No data are available on the toxicity of Amines, C12-14-tert-alkyl, mixed sec-Bu and iso-Bu phosphates (CAS No. 96690-34-5) to aquatic algae and microorganisms. Therefore data from the analogue substance Amines, C11-14-branched alkyl, monohexyl and dihexyl phosphates (CAS 80939-62-4) were used as read across. The target and the source substance are expected to have a similar environmental fate and ecotoxicity profile due to the comparable physico-chemical properties of both substances. Both the target and source are UVCB substances comprising amine components and organophosphates.The target substance has C12-14 chain lengths in the amine component, whereas the source has C11-14 alkyl chains. The organophosphate group of the target substance has different butyl phosphate constituents, while the source substance has varying hexyl phosphates.
The target and the source substance are not volatile (VP ≤ 54 Pa and 0.00005 Pa at 20 °C, respectively). The amine constituents of the target substance have estimated partition coefficients ranging from 4.65 to 5.63, while the organophosphate constituents of the target substance have log Kow values ranging from 0.69 to 2.14. Whereas the amine constituents of the source substance have estimated partition coefficients ranging from 5.3 to 11.6, while the organophosphate constituents of the source substance have log Kow values ranging from 1.7 to 4.3. The water solubility of the source substance is 1322 mg/L at 25 °C and pH 3.5. For the target substance water solubilties of 37 mg/L at a loading of 1 g/L and 191 mg/L at a loading of 10 g/L at 20 °C and pH 4.6 were determined. Despite the difference in water solublities both substances share a common ecotoxicological profile as indicated by the available studies for both substances. Thus read across to the source substance is considered justified. This read-across is in accordance with Regulation (EC) No. 1907/2006 Annex XI, 1.5 and justified in detail in the analogue justification attached in IUCLID Section 13.
The available test on the toxicity of the source substance on green algae (Pseudokirchneriella subcapitata) determined an EC50 (72 h)> 10 mg/L and a NOEC (72 h) of 3.2 mg /L (nominal). Testing for inhibition of respiration of activated sludge microorganisms resulted in a NOEC (14 d) >= 100 mg/L. Therefore, it is not anticipated that the test item inhibits the degradation process in sewage treatment plants.
