In the respirometry test the biodegradation of test substance and the toxicity of test substance towards activated sludge microorganisms is examined. In closed flasks, mixtures of activated sludge, nutrient salts, the test substance and a reference substance are incubated for 28 days under controlled conditions. The biochemical oxygen demand (BOD) is measured throughout the test and at the end of the incubation period the dissolved organic carbon content in the test flaks is determined.
A toxicological effect is defined as inhibition of respiration of the activated sludge which is measured as BOD values at t=24h and to account for possible adaptation effects, also at the end of the test. The respiration inhibition in the flasks containing activated sludge, test substance and reference substance is compared to flasks containing activated sludge and reference substance only. - GLP compliance:
