One study evaluating the acute toxicity of Glycerides,C14-18 and C16-18 unsaturated, mono-, di- and tri- (CAS No. 91052-28-7) is available. This test was performed according to the German national Guideline DIN 38412/15 (Examination of water, waste water and sludge; bioassays (L); determination of the effect of substances in water on fish- Fish test (L15)). In this static test, fish (Leuciscus idus) were exposed to the test substance for 48 hours. After this period, no mortality or any other effects were reported at any of the concentrations tested (LC50 (48 h) > 10000 mg/L).

According to the Guidance on information requirements and chemical safety assessment, chapter R.7b, results obtained at shorter test durations that the standard 96 hours (OECD 203) should be treated with caution and always in combination with other data (read-across, non-testing, etc.). Therefore, a Weight of Evidence (WoE) approach is applied to cover adequately this endpoint (in accordance with Regulation EC (No). 1907/2006, Annex XI, 1.2). The available data for Glycerides, C14-18 and C16-18 unsaturated, mono-, di- and tri- are used in combination with an additional study performed on the structurally related category member (1,2,3-propanetriyl trioleate (CAS No. 122-32-7)) in accordance with Regulation (EC) No. 1907/2006, Annex XI, 1.5. Both substances are fatty acid esters with glycerol as alcohol component. They are UVCB substances with a high content (>60%) of C18 unsaturated fatty acid and to a certain extent C16, C14 and C12, and their water solubilities are comparable (< 0.05 mg/L). Therefore, these two substances are expected to show similar behaviour in the water phase and comparable degrees of toxicity to aquatic organisms.

 

The study with 1,2,3-propanetriyl trioleate (CAS No. 122-32-7)(Bogers, 1988) was conducted according to the German Method Evaluation of hazardous substances in water- Determination of the acute toxicity to Fish, under GLP conditions. Fish (Cyprinus carpio) were exposed to the test substance within a static water regime at 23 ± 2°C. During the exposure period, no mortality was observed in treatment or control groups. The LC50 (48 h) was determined to be > 1000 mg/L (nominal concentration).

 

The results from the two 48-h studies combined provide enough evidence to state that Glycerides,C14-18 and C16-18 unsaturated, mono-, di- and tri- is not expected to be acutely toxic to fish up to the limit of water solubility (< 0.05 mg/L).
