Description of key information

N,N-dimetyhbutylamine is acutely toxic to fish. The estimated LC50(96 hr)  is 35 mg/L.

The acute aquatic toxicity of N,N-dimethylbutylamine was estimated with two different QSAR program, i.e. the ECOSAR program (v1.00) that is part of the EpiSuite (v4.10) software package, and with TEST (v4.1). Both QSAR models were developed by the US EPA.

The ECOSAR results indicate that the LC50(96 hr) to fish (fresh water) is approx. 35 mg/L for the free base, and approx. 200 mg/L for the neutral molecule. The LC50(96 hr) estimate obtained with TEST was 64.3 mg/L (GUBDH, 2012). The lower value is selected for further considerations.

An experimental GLP guideline study on N,N-dimethylbutylamine using zebra fish is available. The LC50(96 hr) was 212 mg/L for the neutralized test substance, i.e. the experimental value is in good agreement with the ECOSAR estimate. The dose-mortality curve was very steep sind 10% mortality was seen at 180 mg/L and 90% mortality at 250 mg/L /Markert and Jung, 1988). Anyhow, the estimated an dexperimental data indicate that the free base is much more toxic than the neutralized form.
