- Three days before starting of the test (main test), an algal pre-culture was made which was
inoculated from an algal stock culture derived from the algal type culture collection Göttingen in
order to guarantee sterile conditions at all. The pre-culture was incubated under the same
conditions as the test solutions to be tested afterwards.
At t0 the test solutions were prepared. Absorbance values of the solutions were determined at t0
(theoretical value on basis of the measured value for the inoculum), after 25 hours, 49 hours and
72 hours using a spectrophotometer at 578 nm.
The incubation of the test and control solutions took place according to the OECD Guideline 201
for a period of 72 hours at a temperature of ≈23.5°C at an illumination rate of >120
μE/m2s ([8000 Lux] measured once per month with a spherical collector within the range of 400-
700 nm). In order to assure the input of CO2, the test solutions were stirred by means of a
magnetic stirrer for the duration of 15 minutes per hour. Longer stirring periods were avoided to
prevent damage of the algal cells.

must be at least 16. With the current test it was found to be 189,0. The test fulfills this validity criterion.

- Executive summary:
The aim of this study was the determination of the chronic toxicity towards algae of “308 AQN” according to OECD-Guideline No. 201, in the Version dated 23-March-2006.
The toxic behaviour was investigated by determination of the inhibition of the growth rate of the algae and the yield during the exposure period of 72 hours. As the test item exhibits an extreme poor water solubility it was tested using aqueous extracts (“WAFs”) at nominal concentrations up to and including 1000 mg test item/L (final concentration). The main test was performed as a limit test using “WAF” with nominal concentation from 1000 mg/L of the test item. The following EL-values (“effective loading”) with 308 AQN are assumed:

Supporting chemical analyses were not performed within this study as the test item could not be determined by any analytical method because of the huge molecular weight of the molecule. Based on this, the reported effect concentrations (EL values) should refer to nominal concentrations of the test item. The validity criteria for the study as given by the OECD Guideline 201 were met and the study is therefore considered to be valid.
