Short-term toxicity to aquatic invertebrates
Administrative data
Link to relevant study record(s)
- Endpoint:
- short-term toxicity to aquatic invertebrates
- Type of information:
- experimental study
- Adequacy of study:
- key study
- Study period:
- 48 hours
- Reliability:
- 1 (reliable without restriction)
- Rationale for reliability incl. deficiencies:
- guideline study
- Qualifier:
- according to guideline
- Guideline:
- OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test)
- Deviations:
- yes
- Remarks:
- The temperature within the incubator had a recorded range between 19.5 – 20.7°C and therefore was not maintained within ±1°C throughout the test but was still within the specified range of 20±2°C. No impact on the integrity and validity of the study.
- GLP compliance:
- yes
- Specific details on test material used for the study:
- Identification: ColaLiquid DC-5
Name as supplied: ColaLiquid DC-5
Lot/Batch Number: 53467J16
Purity/Composition: CAS Number: 68855-69-6
Cocamide diisopropanolamine 84-88%
Glycerin 5-8%
Free Amine (DIPA) <5%
Chemex reference: ECO 161203 - Analytical monitoring:
- yes
- Details on sampling:
- Duplicate samples were taken at the start and end of the 72 hour exposure period. Samples were taken from remaining test media after filling test vessels for 0 hours and pooled replicate flasks for 72 hours.
- Vehicle:
- yes
- Remarks:
- Elendt M4 medium
- Details on test solutions:
- Stock solutions were prepared for both the range-finding and definitive tests by addition of the test sample directly to Elendt M4 medium in a volumetric flask of adequate volume. Calculated volumes of the stock solution were added to nutrient growth medium and made to volume to prepare the test concentrations and provide enough solution for testing and subsequent water quality measurements.
The test concentrations for both the range-finding test and definitive test were prepared separately by the addition of the test sample to Elendt M4 medium in volumetric flasks of adequate volume to provide enough solution for testing and subsequent water quality measurements. - Test organisms (species):
- Daphnia magna
- Details on test organisms:
- Source: From cultures maintained at Chemex Environmental International Ltd since July 2014 and originally sourced from a clone of genotype 5.
Date of gravid isolation: 12 March 2018
Age of Daphnia at start of test: ≤ 24 hours.
Culture conditions: Cultured under static conditions
Temperature: 20 ± 2°C
Dissolved oxygen: >60% (or 3mg/l) ASV
Photoperiod: 16 hours light and 8 hours dark
Feeding regime: Each working day, a suspension of Chlorella vulgaris. The Daphnia in the test were not fed. - Test type:
- static
- Water media type:
- other: Elendt M4 reconstituted Daphnia water
- Total exposure duration:
- 48 h
- Test temperature:
- Temperature range within incubator throughout test: 19.5 – 20.7 °C
(Required: 20±2°C maintained to ±1°C) - pH:
- pH range in control and test concentrations throughout test: 6.82 – 7.53
- Dissolved oxygen:
- Initial Dissolved Oxygen at 0 Hours: 8.10mg/l
DO range in control and test concentrations throughout test: 7.90 – 9.30mg/l (Required: ≥3mg/l) - Nominal and measured concentrations:
- Results for the analytical confirmation of exposure concentrations indicate that the measured concentrations remained within 80-120% of nominal (actual values 107.335 – 119.938%). Therefore effect concentrations are reported as nominal concentrations of ColaLiquid DC-5 as received
- Duration:
- 24 h

- Duration:
- 48 h
- Dose descriptor:
- NOEC
- Effect conc.:
- 1.2 mg/L
- Nominal / measured:
- nominal
- Conc. based on:
- test mat. (dissolved fraction)
- Basis for effect:
- mobility
- Details on results:
- The Daphnia were examined at 24-hours and 48-hours for abnormal behaviour during the determination of immobility. Abnormalities were observed during the test period 1 individual in 1.2mg/l and 3 individuals in 2.0mg/l test concentrations at 24 hours showing slower movement
- Results with reference substance (positive control):
- None (0%) of the twenty control Daphnia maintained in dilution water were immobilised and none were trapped in the meniscus during the study. This fulfills the control validity criteria of the study as the percentage of immobilisation did not exceed 10%, confirming that the test organisms were of suitable “health” for use in the study.
- Validity criteria fulfilled:
- yes
- Conclusions:
- The 24-hour EC50 and 48-hour EC50 of ColaLiquid DC-5 to Daphnia magna were 1.87mg/l and 1.6mg/l respectively.
The 0 to 24-hour NOEC and LOEC were 1.2mg/l and 2.0mg/l respectively.
The 0 to 48-hour NOEC and LOEC were 1.2mg/l and 2.0mg/l respectively.
The lowest test concentration which immobilised all twenty Daphnia after 48-hours was 3.5mg/l. Immobilisation occurred was in all test concentrations
