Toxicity to aquatic algae and cyanobacteria

Description of key information

Freshwater algal growth inhibition test was carried out on Desmodesmus subspicatus with the substance Sodium 5-(aminosulphonyl)-2-(7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl)benzoxazolesulphonate according to OECD Guideline 201[ABITEC study report, Sustainability Support Services (Europe) AB (study report no. 515/16, 2016]. The stock solution (200 mg/L) was prepared by dissolving orange powder in OECD growth medium. Test solutions of required concentrations were prepared by mixing the stock solution of the test sample with OECD growth medium and inoculum culture. Conc. of test chemical used for the study was 1.2, 10, 25, 50, 100 and 200 mg/L, respectively. The test was performed under static conditions in a static fresh water system at a temp. of 23±2°C. Initial cell density of test organism used was 5x10(3) cells/ml. Determination of cell counting involve the use of microscope with counting chamber Cyrus I or electronic particle counter. ErC50 was calculated using non-linear regression by the software Prism 4.0. The median effective concentration (EC50) for the test substance Sodium 5-(aminosulphonyl)-2-(7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl)benzoxazolesulphonate in Desmodesmus subspicatus was determined to be 164.2 mg/L on the basis of effects on growth rate in a 72 hour study. Thus based on this value, test chemical Sodium 5-(aminosulphonyl)-2-(7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl)benzoxazolesulphonate can be considered as non-toxic to aquatic organisms and thus cannot be classified as hazardous as per the CLP criteria.

The effect of test item sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate, CAS No. 93859-32-6 was studied on the growth of fresh water green algaChlorella vulgaris.The study was conducted following OECD guideline 201- Alga, growth inhibition test [UERL Study Report, Sustainability Support Services (Europe) AB (Report no. 93856-32-6/01/2016/AT), 2016].The test concentration chosen for the study were 6.25mg/l, 12.5mg/l, 25mg/l, 50mg/l,100mg/l and 200mg/l. The test concentrations were prepared using stock solution of the test item using mineral media. The green alga was exposed to the test concentration for a period of 72 hours to observe average specific growth rate and percentage growth inhibition under the effect of the test item. EC50 calculated graphically through probit analysis was observed to be 40.605mg/l. Thus based on this value, it can be concluded that the substance can be classified as aquatic chronic category 3. Since the partition coefficient (log Kow) of test chemical is not ≥ 4 (i.e, reported as -0.21), test chemical sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate can be considered as non-toxic to aquatic organisms and thus cannot be classified as hazardous as per the CLP criteria.

 

72 hrs aquatic toxicity study was predicted (SSS QSAR Prediction model,, 2016) to assess toxic effects of the test compound Sodium 5-(aminosulphonyl)-2-(7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl)benzoxazolesulphonate (CAS No.93856-32-6) and the results were predicted. The study was based on the effects of the test compound on Desmodesmus subspicatus in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the test compound Sodium 5-(aminosulphonyl)-2-(7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl)benzoxazolesulphonate (CAS No.93856-32-6) was estimated to be211.225 mg/l on the basis of growth rate. Thus based on this value, test chemical Sodium 5-(aminosulphonyl)-2-(7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl)benzoxazolesulphonate can be considered as non-toxic to aquatic organisms and thus cannot be classified as hazardous as per the CLP criteria.

Based on the QSAR prediction done using the EPI Suite ECOSAR version 1.10 (EPI Suite, 2016), the 96 hours EC50 was estimated to be 20440.041 mg/l on green algae for substance Sodium 5-(aminosulphonyl)-2-(7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl)benzoxazolesulphonate with growth inhibition effects. Thus based on this value, it can be concluded that the substance can be considered as non - toxic to aquatic organisms and thus cannot be classified as hazardous as per the CLP criteria.

Based on the overall reported results of various study for target substance, it can be concluded that the substance can be classified as not hazardous as per the CLP criteria.
