Acute toxicity
The acute oral median lethal dose (LD50) of sodium 3-nitrobenzenesulphonate in rat was found to be 5000 mg/kg of body weight. This value indicates that sodium 3-nitrobenzenesulphonate does not exhibits acute toxicity by the oral route.
Irritation / corrosion
In vivo studies of the substance show that the substance is not irritating to skin but has eye irritation potential, and so the substance is classified in category 2 for eye irritation according to the CLP criteria.
Sensitisation
The number of animals sensitized is primarily taken into account in the evaluation. The control animals are used to rule out any substance-induced primary skin irritation. The findings obtained 24 hours after the removal of the patch are taken into account for the determination of the sensitization rate. The evaluation "sensitizing" results if at least 30 per cent of the animals exhibit skin reactions in this adjuvant test. Under the test conditions chosen and following the described results, Ludigol Granulat has a sensitizing effect on the skin of the guinea pig.

DNEL derivation
Sodium 3-nitrobenzenesulphonate is non acute toxic to oral route, shows no irritation effect to skin but have a property of eye irritation, is not genotoxic and is not a developmental or reproductive toxin.
In the absence of local effects following short-term or long-term exposure, no dose-response data are available and a quantitative dose descriptor is not derived. DNEL values for local exposure are therefore not calculated.
In the absence of acute systemic toxicity, no dose-response data are available and a quantitative dose descriptor is not derived. DNEL values for acute systemic effects are therefore not calculated.
A standard approach to deriving DNEL values would be to use the reproductive toxicity dataset and apply assessment factors as described in ECHA guidance documents. The critical endpoint is considered to be the LOAEL of 162 mg/kg bw/d in oral route.
