- Details on study design: HPLC method:
- This method utilises a high performance liquid Chromatograph with UV/VIS detector, reversed phase column and an aqueous mobile phase. An analytical column packed with a commercially available cyanopropyl solid phase containing lipophilic and polar moieties and a moderately polar stationary phase based on a silica matrix was used.
Test item solution:
FAT 40819/A (19.10 mg) was dissolved in water (50 mL) to prepare a stock solution of 382.0 µg/mL. The stock solution (100 µL) was diluted with a solvent mixture of water and acetonitrile (45:55; v/v) to 10 mL to obtain a standard solution of 3.82 µg/mL. The pH value of the test item solution was determined to be 6.8. This test item solution was injected three times.
HPLC Conditions:
- Apparatus: Varian pump ProStar 230, Varian autosampler 9100, Varian UV/VIS Detector ProStar 310, Jones column oven 7990
- Column: LiChrospher 100 CN, particle size 5 µm, 250 mm x 4 mm
- Precolumn: CN
- Mobile Phase: Water / acetonitrile (45:55; v/v); pH 6.4
- Detection: Test item: 254 nm for test item, 210 nm for reference item and for sodium nitrate.

- Details on results (HPLC method):
- - The test item solution was injected three times and the reference item solutions were injected six times. FAT 40819/A was found to elute as one peak at retention times of about 1.00 minute. Sodium nitrate, that was used for the dead time determination, was eluted at a retention time of about 1.02 minutes. The reference item with the lowest Koc was eluted at a retention time of about 2.84 minutes. FAT 40819/A was not retained under the chromatographic conditions of the test and was eluted at the dead time. The Koc of FAT 40819/A was therefore determined to be lower than that one of the lowest reference item acetanilide.
- The log KoC of FAT 40819/A was estimated to be <1.25 which is equal to a Koc value of <18 using the HPLC method. This value indicates that FAT 40819/A will not be absorbed by organic carbon in soil. FAT 408119/A can be classified to be of very high mobility. - Validity criteria fulfilled:
- yes
- Conclusions:
- With a log Koc of <1.25 (equivalent to a Koc value of <18 L/kg) the substance may be considered to be mobile.
- Executive summary:
In a GLP-compliant study, the determination of the adsorption coefficient (Koc) of the test substance was performed according to OECD Guideline 121 using the HPLC method. The test item solution was injected on the HPLC column. The test substance was found to elute as one peak at retention times of about 1.00 minutes. Sodium nitrate, that was used for the dead time determination, was eluted at a retention time of about 1.02 minutes. The test substance was therefore not retained under the chromatographic conditions of the test and was eluted at the dead time. The Koc of the test substance was therefore determined to be lower than that one of the lowest reference item acetanilide and was estimated to be <1.25 which is equal to a Koc value of <18 using the HPLC method. This value indicates that the test substance will not be absorbed by organic carbon in soil and can be classified to be of very high mobility.
