Description of key information

Half-live of ACH (2 g/l) at pH 7 and 23 °C: 4 min. The half-live increases exponential with pH.

Hydrolysis of ACH at 2 g ACH/l Deuteriumoxide was studied in phosphate buffer solutions at pH 6.0, 6.4, 6.86, 7.0 for 24 hours. The experiment was not conducted according to guideline or GLP but is well documented, comprehensible and meets basic scientific principles. The hydrolysis products of ACH (Acetone and HCN 1:1) are well known. The hydrolysis was followed by analysing the concentration of the hydrolysis product acetone for 6-9 times. Acetone concentration was measured by IR spectroscopy (ATR-FTIR). The half-life (lives)/DT50 (50% decline times) of ACH (room temperature) were 55 min at pH 6.0, 31 min at pH 6.4, 5.4 min at pH 6.86, and 4.0 min at pH 7.00. This study is classified acceptable and satisfies the information needs for hydrolysis study though no guideline was used. Due to the highly toxic properties of ACH a standard guideline could not be used.

The ACH half-life of < 10 minutes at pH 7 is in compliance with the information (validity not assignable) given by AEGL committee (US-NAC, Actone Cyanohydrin, Interim Acute Exposure Guideline Levels (AEGLs), Interim final draft, 2005) and ECETOC (JACC report No. 53, Volume I, 2007).
