Short term toxicity
Reliable acute toxicity tests results are available for freshwater fish (Oncorhynchus mykiss), invertebrates (Daphnia magna) and algae (Pseudokirchnerella subcapitata). LC50, EC50 and ECr50 values for the three organisms were 0.48, 0.14 and 2.1 mg/L, respectively.
In a reliable 96-h acute toxicity study Rainbow trout (Oncorhynchus mykiss) were exposed to 2-butyloctan-1-ol at measured concentrations of 0 (control), 0.32, 0.71, 1.1, 1.8 and 4.2 mg/L under semi-static conditions following OECD 203 guidelines. The 96-h LC50was 0.48 mg/L, based on mortality. Consequently, this value will be taken as the short-term toxicity in fish endpoint.
In a reliable 48-h acute toxicity study Daphnia magna were exposed to 2-butyloctan-1-ol at nominal concentrations of 0 (control), 0.046, 0.1, 0.215, 0.46, 1.0, 3.0, 10, 30 and 100 mg/L under semi-static conditions following OECD 202 guidelines. The 48-h EC50was 0.14 mg/L, based on immobilization. Consequently, this value will be taken as the short-term toxicity in invertebrates endpoint.
In a reliable 72-h toxicity study, the green algae, Pseudokirchneriella subcapitata, were exposed to 2-butyloctan-1-ol at time-weighted mean measured concentrations of 0 (control), 0.058, 0.17, 0.57, 1.5 and 4.3 mg/L under static conditions following OECD 201 guidelines. The 72-h EC50 was 2.1 mg/L based on growth rate. Consequently, this value will be taken as the short-term toxicity in algae endpoints.
In waste-water treatment plant micro-organisms, the structurally related 2-octyldodecan-1-ol does not cause inhibition at the limit of water solubility. In view of the ready biodegradability, further information is not required in REACH.
Long term toxicity
In accordance with Column 2 of REACH Annex IX, the long-term aquatic toxicity to fish study (required in Section 9.1.6) does not need to be conducted as the chemical safety assessment according to Annex I indicates that this is not necessary.
A reliable 21 day NOECrepro value of 14 µg/L was determined for the effects of the structurally related dodecan1-ol on reproduction (cumulative number of offspring) of the freshwater test organism Daphnia magna. This study reflects the only value that is available for this endpoint.

In accordance with Column 2 of REACH Annex X, long-term toxicity testing with sediment organisms (required in Section 9.5.1) is not needed as the chemical safety assessment according to Annex I indicates that this is not necessary.
