Test guideline
- Qualifier:
- according to guideline
- Guideline:
- EU Method C.2 (Acute Toxicity for Daphnia)
- GLP compliance:
- yes
Test material
- Test material form:
- solid: particulate/powder
- Details on test material:
- Batch number: 022401
Constituent 1
Sampling and analysis
- Analytical monitoring:
- yes
- Remarks:
- The actual concentrations of the test substance in the test media were determined by HPLC before the test and after 48 hours.
Test solutions
- Vehicle:
- no
- Remarks:
- Immediately before the beginning of the test, a 100 mg/l solution was prepared by dissolving 25.0 mg test substance in 250 ml reconstituted water. The lower concentrations were then obtained by diluting with reconstituted water.
- Details on test solutions:
- Identity and concentration of auxiliary solvent for dispersal: The test substance was dissolved in dilution water.
Test organisms
- Test organisms (species):
- Daphnia magna
- Details on test organisms:
- Neonates of Daphnia magna, less than 24 hours old, were obtained by hatching ephippia, supplied in the microbiotest "Daphtoxkit F TM "(Daphnia rnagna; MicroBioTests Inc., lndustriezone "De Prijkels", Venecoweg 19 , 9810 Nazamth, Belgium). Batch No. of the ephippia: M291002. The neonates were collected for the test at 86 hours after initiation of hatching.
Study design
- Test type:
- not specified
- Water media type:
- other: Immediately before the beginning of the test a 100 mg/l solution was prepared by dissolving 25.0 mg test substance in 250 ml reconstituted water. The lower concentrations were then obtained by diluting with reconstituted water.
- Total exposure duration:
- 48 h
Test conditions
- Hardness:
- 250 mg CaCO3/L
- Test temperature:
- Temperature: approximately 21 °C throughout the test.
- pH:
- pH: between 7.5 and 8.5 in the test substance solutions.
- Dissolved oxygen:
- Dissolved oxygen concentration: at least 8.3 mg/L.
- Nominal and measured concentrations:
- Nominal concentrations (mg/L): 0, 10, 18, 32, 56, 100.
At the end of the test the measured concentrations of "STl571 F8" were between 96 and 101 % of the actual starting concentration.

- Details on results:
- At the end of the test the measured concentrations of "STl571 F8" were between 96 and 101 % of the actual starting concentration.
Immobilisation of control group daphnids was O % at the end of the test (guideline: maximum of 10 %).
As some daphnids in the test substance groups wem trapped at the water surface there is a possible influence of the test substance to the water surface tension.
There were no control daphnids trapped at the water surface.
Temperature was stable during the test period.
Dissolved oxygen concentration in the test vessels was not lower than 8.4 mg/L (EC guideline: higher than 3 mg/L).
Applicant's summary and conclusion
- Validity criteria fulfilled:
- yes
- Remarks:
- Dissolved oxygen concentration in the test vessels was not lower than 8.4 mg/L (EC guideline: higher than 3 mg/L).
- Conclusions:
- Acute aquatic toxicity: Cat.3
