Test solutions

Vehicle:
yes
Remarks:
reconstituted water
Details on test solutions:
The test medium was prepared as follows:

prior to the test start, a 10.0 mg test item/L stock solution was prepared by direct weighting of 0.0050 g of test item into 500 mL of reconstituted water. The obtained solution appeared clear with particles if test item. This solution was sonicated for about five minutes and after was stirring with magnetic stirrer for about 2 hours. After stirring the stock solution appeared clear and transparent.

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
A strain of Daphnia magna Straus was supplied by MicroBioTests Inc., Belgium in July 2017 (batch number: DM060417).
Since that time, the clone has been bred in the laboratories of the Test Facility under conditions identical to those of the test, with regard to temperature, light and water quality, so that no acclimation was needed prior to testing.
It was daily fed with a fixed amount of the laboratory cultured green algae Pseudokirchneriella subcapitata (formerly Selenastrum capricornutum) and with a suspension of the yeast Saccharomyces cerevisiae.
At the test start the organisms used in the test were less than 24 hours old and were not first brood progeny.

For the test concentration and for the negative control, group of 20 daphnids were used divided into four replicates of five daphnids each. The daphnids were randomly distributed to the test vessels at test initiation. The daphnids loading rate was about 8 mL per one daphnid

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
A full test at five concentrations, 0.4, 0.9, 2.1, 4.6 and 10.0 mg test item/L, was performed to evaluate the effect of the test item on Daphnia magna under static conditions.

The daphnids were observed for immobility after 24 and 48 hours of exposure.
EC0, EC100, EC50 (with their confidential limit) and NOEC, expressed in terms of nominal test item concentration, were as follows:

Time (h) EC0 (mg/L) EC100 (mg/L) EC50 (mg/L) NOEC (mg/L)
24 0.9 4.6 2.2 (1.9 – 2.7) 0.9
48 0.4 4.6 1.0 (0.8 – 1.2) 0.4
