- Mixed Inoculum Preparation:1 capsule of polyseed was dissolved into 500ml DI water and stirred it for 1 hour for proper mixing. This gave the
bacterial count as 107 to 108 CFU/ml. At the regular interval microbial plating was also performed on agar to confirm the vitality and CFU count of

- 1. The test item, i.e., 4,4'-carbonimidoylbis[N,N-diethylaniline] monohydrochloride was taken at a concentration of 0.5ml/125ml The
reference item chosen was taken at a concentration of 0.5ml/125ml in the mineral medium was directly added to the BOD bottle.
2. The BOD bottle of test item& reference item were filled with 120ml &120.5ml of mineral media, respectively.
3. All the BOD bottles were inoculated with prepared mixed inoculum (Polyseed) at the concentration of 32ml/L.. This concentration gave the
bacterial CFU/ml as 107to 108.
4. Analyse all the zero-time bottles immediately for dissolved oxygen by the electrode methods. Stopper the remaining replicate bottles ensuring
that no air bubbles are enclosed, and incubate at 20°C in the dark.
5. Each series must be accompanied by a complete parallel series for the determination of the inoculated blank medium.
6. Withdraw at least duplicate bottles of all series for dissolved oxygen analysis at time intervals (at least weekly) over the 35 days incubation.
7. For N-containing test substances, corrections for uptake of oxygen by any nitrification occurring should be made. - Reference substance:

Accordingly, the % degradation of the test item after 35 days of incubation at 20 ± 1°C according to Closed Bottle test was found to be 22.51%. Based on the results, the test item, under the test conditions, was considered to be primary inherent biodegradable.
