Sampling and analysis
- Analytical monitoring:
- yes
- Details on sampling:
- - Sampling method: At the start of the exposure (0 h) samples from vessels without daphnids and at the end of the exposure (48 h) samples from vessels with daphnids were analysed. The samples were transported to the Analytical Laboratory on the day of sampling.
Test solutions
- Details on test solutions:
- PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Prior to use the test substance was homogenized with a pestle. For correct dosing of the test substance acetone was used as solvent. For each test concentration the appropriate volume of the acetonic solution of 50 mg test substance in 500 mL acetone was prepared. The acetonic solution was distributed on the bottom of the test vessel to maximize the surface and the acetone was completely evaporated. After this 400 mL test medium were added and the test vessels were shaken for about one day at the test temperature (at 50 rpm). The pH of the test solutions was not adjusted.
To ensure that the organisms were only exposed to dissolved test substance, the exposure was carried out in glass beakers into which were placed organism chambers consisting of a glass tube with a stainless steel mesh bottom. The glass tubes were submerged in beakers with test medium with the top of the tube sufficiently above the surface of the test medium. The bottom of the tube was suspended above the bottom of the test vessel preventing direct contact with the layer of undissolved test substance.
