Propane-1,2,3-triyl 3,5,5-trimethylhexanoate (CAS No. 56554-53-1) is not expected to be acutely or chronically toxic to aquatic organisms in the range of water solubility. Experimental data on the acute toxicity of propane-1,2,3-triyl 3,5,5-trimethylhexanoate (CAS No. 56554-53-1) are available for fish and algae. The short-term toxicity of the substance to Zebra fish (Danio rerio) was tested in a GLP guideline study according to OECD guideline 203 (LC50 (96 h) >1.0 mg/L (nominal), correpsonding to 100% v/v. Thus a toxic effect of the substance was not observed up to the limit of water solubility (< 0.05 mg/L). The toxicity of the test item to green algae Raphidocelis subcapitata was assessed, according to GLP and OECD 201, as a limit test. Acute effects on the algal growth were not observed. Based on the geometric mean measured test concentration an ErC50 (72 h) of greater than 0.0075 mg/L was determined.
Furthermore, in this test no chronic effects on algae were observed, resulting in a NOErC (72 h) of ≥ 0.0075 mg/L (geometric mean measured concentration). The long-term toxicity of the substance to Daphnia magna was tested in a guideline study following OECD 211. An effect on survival or reproduction rate of the daphnids was not observed within the limit of water solubility (NOEC (21 d): ≥1.0 mg/L (nominal), correpsonding to 100% v/v). The long-term toxicity of the substance to eggs and sac-fry stages of Pimephales promelas was tested in a guideline study following OECD 210. An effect on survival, hatching rate or growth of the fish was not observed within the limit of water solubility (NOEC (33 d): ≥0.081 mg/L (geometric mean measured concentration).
Data on the toxicity to microorganisms was derived from the toxicity control of the biodegradation study according to OECD 301B. A respiration inhibition of activated sludge was not observed and thus the substance concentration of 14.2 mg/L was used as NOEC. In conclusion, propane-1,2,3-triyl 3,5,5-trimethylhexanoate (CAS No. 56554-53-1) shows no acute and chronic toxic effects on freshwater organisms up to the limit of the water solubility of the test substance.
