In a study performed by Barua (2012) the effects of the test substance on chemokine and cytokine production in macrophages was evaluated. In addition, changes in intracellular signaling pathways and activation of specific gene promoters was assessed. RAW 264.7 mouse macrophages were treated with the test substance and examined for changes in immune function. Luminex multiplex technology was used to assess changes in cytokine/chemokine expression and activation of kinase signalling pathways. In addition, a panel of macrophage cell lines with promoter-specific luciferase reporter genes were generated and treated with the test substance, and transcriptional responses to these chemicals were detected by changes in luminescence. The production of IL-1β, TNF-α and G-CSF were >50, >50 and > 25 % increased relative to untreated control, respectively. IL-1α and IL-6 showed a >50% suppression relative to the untreated control and MIP-1β, RANTES showed a >25% suppression relative to the untreated control. No changes were observed for MCP-1, MIP-1α, and MIP-2. The following intracellular signaling pathway changes were observed: the activation state of CREB, Erk–MAP kinase 1/2, p38, STAT3 were induced >25% relative to untreated control. No changes were observed in IκB-α, JNK, p70 S6 kinase, STAT5A/B. The following changes in gene expression were observed: SRE was induced >20% and CRE, RBP/Jk, and TCF were >25% suppressed relative to untreated control. No changes were observed in the gene expression of AP1, C/EBP, GAS, ISRE, NFAT, NFkB, SMAD, STAT3.
