Mordant Black 9 was negative in the Ames Test with Prival modification, tested in 4 stains of Salmonella typhimurium.

The structural analogue Diamantechtrot BT was tested in a Bacterial mutation test in 4 Salmonella and one E. coli strain in the standard Ames and the Prival modification. In the presence of hamster liver S9-mix (Prival modification) using the preincubation method according to Prival Diamantechtrot BT resulted in significant and dose-dependent increases in the number of revertant colonies with the tester strains TA 1535 and WP2uvrA.

Due to this test result, the potential of Diamantechtrot BT to induce gene mutations at the HPRT locus in V79 cells of the Chinese hamster in vitro was tested.Up to the highest investigated dose no relevant and reproducible increase in mutant colony numbers was obtained in two independent experiments. Hence, Diamantechtrot BT does not induce gene mutations in the HPRT-test with V79 Chinese hamster cells, both in the presence as well as in the absence of a metabolic activation system, under the experimental conditions described. Diamantechtrot BT is therefore considered to be non-mutagenic.

In addition, Diamantechtrot BT was tested for its potential to induce chromosome aberrations in V79 cells of the Chinese hamster lung in vitro and was found to be non-mutagenic in this chromosome aberration assay.
