Test solutions

Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION
The test substance was pipetted carefully, on the water surface outside of the glass tubes. The test vessels were covered with Parafilm “M” and shaken slowly for about one day, so that the test substance did not form an emulsion. Before the organisms were introduced into the glass tubes the surface in the glass tubes was checked for undissolved substance. Throughout the exposure period oily drops of undissolved test substance were observed on the water surface outside of the glass tubes, while no test substance was visible on the water surface in the glass tubes.

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
- Common name: Daphnia magna STRAUS
- Source: Daphnia magna STRAUS was originally supplied from "Institut National de Recherche Chimique Appliquée, France", and has been breeded from 1978 in the Laboratories of BASF.
- Age at study initiation: 2 - 24 hours old

Study design

Test type:
static
Water media type:
freshwater
Limit test:
yes
Total exposure duration:
48 h

Test conditions

Hardness:
2.48 mmol/L
Test temperature:
20.2 - 21.0 °C
pH:
8.3
Dissolved oxygen:
8.8 - 8.9 mg/L
Nominal and measured concentrations:
Nominal concentration: 100 mg/L, a control was tested in parallel.
Details on test conditions:
TEST SYSTEM
- Test vessel:
- Type: open
- Material, size, headspace, fill volume: glass beakers (nominal volume 600 mL) into which organism chambers were placed (cylinders of transparent glass, 6 cm diameter, 10 cm high, with a bottom of stainless-steel grid of 0.2 mm x 0.2 mm). The daphnia were placed in the organism chamber, to prevent direct contact of the test organisms with undissolved test substance on the surface of the test solution during the test. 400 mL total volume in the glass beakers.
- No. of organisms per vessel: 5
- No. of vessels per concentration (replicates): 4
- No. of vessels per control (replicates): 4

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: M4 medium with the properties:
Total hardness: 2.20 - 3.20 mmol/L
Alkalinity up to pH 4.3: 0.8 - 1.0 mmol/L
Ratio Ca/Mg ions: about 4:1
pH value: 7.5 - 8.5
Conductivity: 550 - 650 µS/cm

OTHER TEST CONDITIONS
- Photoperiod: 16:8 hours, diffuse light (20 – 850 Lux at a wave length of 400 - 750 nm)

EFFECT PARAMETERS MEASURED :
Observations: mobile daphnids after 0, 24 and 48 h
Measurements: Temperature continuously during the whole test period in a separate vessel proximal to the test vessels, pH and oxygen content after 0 and 48 h
Reference substance (positive control):
yes
Remarks:
Potassiumdichromate

Results and discussion

Effect concentrations
Duration:
48 h
Dose descriptor:
EC50
Effect conc.:
> 100 mg/L
Nominal / measured:

Basis for effect:
mobility
Details on results:
Mortality of control: no
In conclusion the 48 hour EC50 value for C16+ - Olefingemisch in the Daphnia magna study was > 100 mg/L based on the loading rate of the test substance. No effect was observed up to the solubility limit of the test substance in water under the test conditions.
Results with reference substance (positive control):
EC50 (24 h) = 1.04 mg/L
