- 1000 mg of test material was dispersed in culture medium with the aid of ultrasonication for approx. 30 minutes and the volume adjusted to 500 mL to give a 2000 mg/L dispersion.
- Additional stock solutions were prepared by serial dilution.
- An aliquot of each of the stock dispersions was mixed with algal suspension (250 mL) to give the test concentrations: 62.5, 125, 250, 500 and 1000 mg/L.

GROWTH RESULTS:
- Both growth rate (r) and biomass (b) were affected by the presence of the test substance.
- For the area under the growth curve, there were no statistically significant differences between the control, 62.5, and 125 mg/L test concentrations ( P ≥ 0.05).
- The concentration of the control cultures increased by a factor of 71 (acceptance criteria is a factor of 16).
- The standard test method indicates that a re-growth study should be done to determine whether toxicity was due to the test substance or due to a reduction in light intensity. Due to a technical oversight, the re-growth study was not conducted. However, spectrophotometer readings indicated that the TS significantly absorbs light at 460 and 665 nm, the wavelengths required for photosynthesis, and therefore, it is probable that the observed toxicity of the TS was due to a reduction in light intensity and not the intrinsic toxic properties of the test material.

OBSERVATIONS ON CULTURES:
- All test and control cultures were observed microscopically at 72 h; misshapen cells were observed at 1000 mg/L.

OBSERVATIONS ON TEST MATERIAL SOLUBILITY:
- At test start, all control cultures were observed to be clear, colorless solutions; at 62.5 mg/L, the cultures were pale brown dispersions; at 125, 250, and 500 mg/L, the test cultures were observed to be brown dispersions, while the 1000 mg/L cultures were black/brown dispersions.
- After 72 h, the 1000 mg/L cultures were black/brown dispersions, and all other cultures were brown dispersions.

PHYSICO-CHEMICAL MEASUREMENTS:
- The temperature was maintained at 20 +/- 1 deg C.
- The pH range was 8.0-8.1 (determined at 0 and 72 h).

POSITIVE CONTROL:
- EbC50 (72 h): 1.1 mg/L, 95% CI = 0.96 - 1.3 mg/L

Reported statistics and error estimates:
One way ANOVA incorporating Bartlett's test for homogeneity and Dunnett's multiple comparison procedure for comparing several treatments with a control was carried out on the area under the growth curve data at 72 hours for the control and all test concentrations to determine any statistically significant differences between the test and control groups. EC50 values were determined from the equation for the fitted line in a plot of percent inhibition versus concentration.
