Test guideline
- Qualifier:
- according to guideline
- Guideline:
- OECD Guideline 201 (Alga, Growth Inhibition Test)
- GLP compliance:
- yes
Test material
- Reference substance name:
- Nickel, compound with Titanium (1:1)
- EC Number:
- 610-765-8
- Cas Number:
- 52013-44-2
- Molecular formula:
- NiTi
- IUPAC Name:
- Nickel, compound with Titanium (1:1)
- Details on test material:
- 100% Equiatomic NiTi intermetallic compound
Batch 1048
Constituent 1
Sampling and analysis
- Analytical monitoring:
- yes
Test solutions
- Details on test solutions:
- The test item has been prepared as a stck solution (10 g/l) and then this solution and 4 different concentrations in water (max solubility) have been tested. Because the test item is poorly soluble in water to facilitate its solubility in water the stock solution has been stirred for 70 hours and then before the beginning of the test this solution has been ultrasonicated for 30 minutes.
The total volume of the culture medium and the concentration of the algae inoculum added are the same in all vessels.
Test organisms
- Test organisms (species):
- Pseudokirchneriella subcapitata (previous names: Raphidocelis subcapitata, Selenastrum capricornutum)
- Details on test organisms:
- TEST ORGANISM
- Family: chlorophyceae
- order: chlorococcalles
- culturing: the algal suspension has been kept in culture medium in sterile 250 ml Erlenmeyer flasks at 23 +/- 2 °C, with lighting greater than 4300 lux and steady with electrical stirrer
Study design
- Test type:
- not specified
- Water media type:
- freshwater
- Limit test:
- yes
- Total exposure duration:
- 72 h
- Post exposure observation period:
- Every 24 hours from the beginning of the test until the end of the test (72 hours), cellular concentration has been determined, by means of Burker chamber, of treated and of control organisms.
Test conditions
- Test temperature:
- 23 +/- 2 °C
- pH:
- from 7.70 to 9
- Nominal and measured concentrations:
- Administered solutions (g/l): 10.00, 3.130, 0.980, 0.310, 0.095
- Details on test conditions:
- Because the test will done on the extract of the test item, constituted by metals with a low volatilisation from water to air, at the beginning and at the end of the test have been checked the concentrations of two metals (nichel and titanium) in the extract by ICP-OES.
Results and discussion
Effect concentrations
- Duration:
- 72 h
- Dose descriptor:
- EC50
- Effect conc.:
- > 10 g/L
- Conc. based on:
- test mat.
- Basis for effect:
- growth rate
- Details on results:
- A normal and healthy appearance of the algae at the end of the test was observed at microscopic.
