- Stock solution of the test substance (1000 mg/L): adding 0.10 g of the test substance to 100 mL of the test water, and then facilitating its dispersion by ultrasonication for 30 min.

All organisms used for a particular test originated from cultures established from the same healthy stock of daphnia. The stock animals were maintained in culture conditions (light, temperature, and medium) similar to which used in the test. - Test type:

TEST MEDIUM:
Good quality tap water which had been dechlorinated for at least 24 h was used. The total hardness of the test water was 122.2 mg (CaCO3)/L, oxygen concentration was about 98% of the air saturation value and pH was 7.55 at room temperature. Characteristics of the dilution water are measured at least twice a year by Jiangsu Provincial Center for Disease Prevention and Control.
RANGE FINDING:
The range-finding test, carried out under static conditions, was regularly conducted to determine the range of concentrations for the following test.
In the range-finding test, groups of animals (5 per group) were exposed to at 3 concentrations of 1, 10 and 100 mg/L (nominal concentration). Synchronously one control group was used in the test. For each test, five daphnia were randomly assigned to 50 mL different concentrations of test substances. - Executive summary:
According to “The guidelines for the testing of chemicals” (HJ/T 153 -2004) and with reference to Procedure 201of the “Guidelines for Testing of Chemicals” of the OECD: “Daphnia sp., Acute Immobilisation Test” (2004) etc., this test was designed to determine the acute toxicity of test substance(meta-Diisopropanolbenzene) to Daphnia magna. The study comprised at least one range-finding test followed by a limit test to determine the 48-hour median effective concentration (EC50) of the test substance. In addition, the NOEC (no observed effect concentration) was also determined.
During the whole test period, the pH values of the control mediums and test mediums were between 7.12 and 7.65, and the Dissolved Oxygen (DO) values varied from 86% to 102% of the air saturation. During the test, the temperature of the test mediums were maintained in the range of 20.1oC to 20.8oC, and all daphnia in the control group were normal. With the same conditions, K2Cr2O7 was used as the negative control substance, and the resulting 24 h-EC50was0.65 mg/L. So the study met the acceptability criteria and was considered valid.
Under valid test conditions mentioned above, the 48h EC50is greater than 150 mg/L (nominal concentration). And 48h NOEC value (no observed effect concentration) is greater than 150 mg/L (nominal concentration). According to “The guidelines for the hazard evaluation of new chemical substances (HJ/T 154-2004)”, the test substance of meta-Diisopropanolbenzene belongs to low toxic chemical for Daphnia magna.
