Executive summary:

In this study the yeast two-hybrid system was used to evaluate the estrogenic activity of hydroxy-anthraquinones. Estrogen agonist and antagonist tests were carried out with or without rat liver S9.

A receptor-binding assay, a Michigan Cancer Foundation-7 (MCF-7) cell proliferation assay (estrogen screen, E-screen), and reporter gene assays in animal cell lines was used to evaluate estrogenic activity.

1,2-dihydroxyaanthraquinone showed toxicity to yeast especially with rat liver S9 than with no S9. In this estrogenicity assay system with the yeast two-hybrid system 1,2-dihydroxyaanthraquinone showed estrogen agonist activity.
