Harmonised classification in accordance with Annex VI of Regulation (EC) No 1272/2008 (CLP Regulation).

Enzymes possess respiratory sensitization potential regardless of the catalytic activities. Enzymes must therefore be classified as “H334: Hazard Category 1: May cause allergy or asthma symptoms or breathing difficulties if inhaled” in accordance with the CLP Regulation. The threshold concentration for classification and labelling with pictogram is 1 % (per individual enzyme), and for allergy warning is 0.1%, based on active enzyme protein (aep).

According to Guidance for identification and naming of substances in REACH (Section 4.3.2.3), enzyme substances are identified by:

a.    the catalytic activity of the enzyme protein (IUBMB nomenclature; active enzyme protein) and

b.    the other constituents from the fermentation (other proteins + peptides and amino acids; carbohydrates; lipids; inorganic salts).

Active enzyme protein (aep): The active enzyme protein (aep) is a potential respiratory sensitizer characterized by the catalytic activity as the main identifier

In general, each individual protein within the “Other proteins” constituent will not be present in concentrations above 1 %.

This constituent is mainly composed of denatured and inactivated enzyme proteins as well as peptides and amino acids. These are not a risk factor under the exposure conditions of industrial enzymes.  

Carbohydrates,  Lipids and Inorganic salts

The carbohydrates, lipids and inorganic salts which are present in the dry matter of an enzyme concentrate are not respiratory sensitizing substances.
