Biodegradation in water: screening tests

The study for the test compound Benzenamine, N-phenyl-, styrenated (CAS No. 68442-68-2) alongwith its predicted data and read across substance wasreviewed for the biodegradation end point which was summarized as below:

Estimation Programs Interface Suite (EPI suite) was run to predict the biodegradation potential of the test compoundBenzenamine, N-phenyl-, styrenated(CAS No. 68442-68-2). The screening test inherent to the biodegradability of the substance was calculated using the software BIOWIN v4.10. The results indicateBenzenamine, N-phenyl-, styrenatedwas estimated to be not readily biodegradable in water.

 

Study (SIDS DOSSIER ON THE HPV PHASE CHEMICAL - Benzenamine, N-phenyl-, styrenated, HPVIS, 2001)was conducted to check the biodegradability status of Benzenamine, N-phenyl-, styrenated. The study was performed according to the OECD Guideline 30 C, modified according to EEC Round-robin-test "Assessment of Biodegradability of Chemicals in Water by Manometric Respiratory DGX 1/283/82 Rec. 5 EEC Directive 79/831 Annex V Part C".

 

The biodegradation was found to be 9 % in 28 days .Therefore the substance may be considered as not readily biodegradable.

 

 

Biodegradation screening test (J-CHECK) was conducted for 28 days for evaluating the percentage biodegradability of the read across substance. Concentration of inoculum i.e, sludge is 30 mg/l and initial test substance conc. used in the study is 100 mg/l. The percentage degradation of test substance was found to be 0% by BOD, and 1% by HPLC. Thus, the substance4-(1-methyl-1-phenylethyl)-N-(4-(1-methyl-1-phenylethyl)phenyl)-is non-biodegradable in water.

5-days Closed bottle test to determine biological oxygen demand and thereby % degradation during the test period was conducted for the read across substanceN, N- diethylaniline(CAS No.91-66-7)[UERLStudy Report, Sustainability Support Services (Europe) AB (Report no.UERL/BOD-005/91-66-7/2014), 2014].

The test system included control, test substance and reference substance. The concentration of test and reference substance (sodium acetate) chosen for the study were 2 mg/l & 5 mg/l respectively while that of inoculum (diluted soil suspension) was 0.5 ml/l. Further, ThOD (Theoretical oxygen demand) of test substance was determined by calculation/experimentally. % Degradation was calculated using the values of BOD and ThOD for test substance and reference substance.

The BOD5value of N,N- diethylaniline(CAS No.91-66-7)was observed to be 0.375 mgO2/mg. ThOD was determined by calculation as 2.78 mgO2/mg. Accordingly, the % degradation of the test substance after 5 days of incubation at 20°C according to closed bottle test was found to be 13.48%. Based on the results, the test item, under the test conditions, was found to be inherently biodegradable at 20°C over a period of 5 days.

 

 

In 2 weeks study (J-CHECK database, 1977) of biodegradation of N,N-diethylaniline, Concentration of inoculum i.e, sludge is 30 mg/l and initial test substance conc. used in the study is 100 mg/l. The percentage degradation of test substance was found to be 0% by BOD, and 1% by HPLC. Thus it is concluded that substance N,N-diethylaniline is found to be not readily biodegradable under test conditions.

 

28-days Manometric respirometry test following the OECD guideline 301Fto determine the ready biodegradability of the test item Bis (4-(1, 1, 3, 3-tetramethylbutyl) phenyl) amine(CAS No. 15721-78-5)[UERLStudy Report, Sustainability Support Services (Europe) AB (Report no.15721-78-5/01/2015/RBD), 2016].

 

The test system included control, test item and reference item. The concentration of test and reference item (Sodium Benzoate) chosen for both the study was 100mg/L, while that of inoculum was 10ml/l. ThOD (Theoretical oxygen demand) of test and reference item was determined by calculation. % Degradation was calculated using the values of BOD and ThOD for test item and reference item.

The BOD28value of Bis (4-(1, 1, 3, 3-tetramethylbutyl) phenyl) amine (CAS No. 15721-78-5) was observed to be 0.0425 mgO2/mg. ThOD was calculated as 3.089mgO2/mg. Accordingly, the % degradation of the test item after 26 days of incubation at 20 ± 1°C according to manometric respirometry test was found to be 1.38% . Based on the results, the test item, under the test conditions, was found to be Non-biodegradable at 20 ± 1°C over a period of 28 days.

 

Biodegradation in water and sediment: simulation tests:

Half life period of Benzenamine, N-phenyl-, styrenated in water is observed to be 37.5 days (900 hrs.) while in sediment it is 337.5 days (8100 hrs). Based on these half life values of Benzenamine, N-phenyl-, styrenated, it is concluded that the chemical is not persistent in water and persistent in sediment.

Biodegradation in soil:

EPI Suite has estimated that Benzenamine, N-phenyl-, styrenated is expected to be found not predominantly in soil and it’s not persistence estimate is based on its transformation in this medium. Its half-life in soil, 75 days (1800 hrs), does exceeds the threshold of 120 days as per Annex XIII of REACH.
