In an acute toxicity test with Daphnia magna the EC50 after 48 h was 3.52 mg/L, the EC50 after 24 h was >4.19 mg/L the EC10 after 48 h was 2.68 mg/L and after 24 h >4.19 mg/L.

Key study:
ln the acute immobilization test with Daphnia magna (STRAUS), the effects of the test item were determined according to OECD 202 (2004) and Council Regulation (EC) No 440/2008 Method C.2 (2008). The study was conducted under semi-static conditions over a period of 48 h with 5 concentration Ievels of the test item. The concentration range was 0.625 to 10.0 mg/L, prepared with dilution water in a geometric series with a factor of 2. Twenty daphnids were exposed to each concentration Ievel and the control. The concentrations of the test item were analytically verified by HPLC-DAD in the fresh media (0 and 24 h) and in the old media (24 and 48 h) of all concentration Ievels and the control. The measured concentrations of the test item in the fresh media (0 and 24 h) were in the range of 41 to 76 % of the nominal values. The measured values in the old media (24 and 48 h) were in the range of < LOQM to 47 % of the nominal values. Therefore, the endpoints were assessed based on the geometric mean measured concentrations of the test item. The geometric mean measured concentrations were calculated to be: 0.156- 0.292- 0.909- 1.54- 4.19 mg/L. The mean recovery rates were in the range of 23 to 42 % of the nominal values. Additionally to the test item another compound with a relative retention time of approx. 0.35 (absorption maximum 291 nm) was detected in all exposure concentrations of the old media. These data show that the area of the metabolite increases with increasing exposure concentration of the test substance. The EC50 after 48 h was 3.52 mg/L, the EC50 after 24 was >4.19. The EC10 after 48 h was 2.68 and after 24 h >4.19.
Supporting study
The acute toxicity of the test item to the water flea Daphnia magna was investigated in a 48-hour static test according to EEC Directive 79/831/EEC, Annex V, Part C: Methods for the determination of ecotoxicity, Ca: Acute toxicit for Daphnia; Updating Nov. 1989. The test substance was tested in the following range of concentrations: 100 to 0.78 mg/L. The dilution factor was 2. Instead of mortality the test criterium of the acute toxicity is immobilization of the daphnids after 48 hours of exposure which expressed as EC (effect concentration). Those animals were considered to be immobile which were unable to swim within 15 seconds after gentle agitation of the test vessel. The following values were determined:
EC0 (48h): 0.78 mg/L
EC 50 (48 h): 3.66 mg/L
EC 100 (48 h): 12.5 mg/L
