- The test medium was prepared by mixing 10 mL stock solution 1 and 1 mL each stock solutions 2, 3 and 4 with 500 mL of sterilized water and then making up the volume to 1 L using sterilized water. The pH of the medium was between 7.82 and 7.83 during the study. This medium was equilibrated with carbon dioxide by keeping it in laminar hood for two hours.

The effect of AGENT 5088-127A was tested on the growth of freshwater single cell green algaPseudokirchneriella subcapitata.
Range finding test was carried out to select the test concentrations for the definitive test. In the range finding test, the tested concentration was 0.00001, 0.0001, 0.001, 0.01, 0.1, 1, 10 and 100 mg/L with single replicate. The test item exhibited1.95, 2.61, 13.68, 17.59, 53.09, 84.04, 92.51 and 100% reduction in cell biomass at the tested concentrations of 0.00001, 0.0001, 0.001, 0.01, 0.1, 1, 10 and 100 mg/L at 72 h.
The alga was exposed to the test item at the nominal concentrations of 0.069, 0.207, 0.621, 1.863, 5.589, 16.767 and 50.301 mg/L (factor of about 3) along with a negative control and a reference substance (Potassium dichromate) at 0.9 mg/L. The cell growth was measured at 24, 48 and 72 hours after the initiation of the test.
The concentration/effect relationship was determined using two factors namely growth rate and yield at the end of the test (72 hours). The per cent inhibition of growth rate was 0.27, 4.92, 19.98, 42.69, 69.65 and 100% and the percent yield inhibition was 1.12, 19.25, 58.21, 85.05, 96.23 and 100% at the nominal concentrations of 0.069, 0.207, 0.621, 1.863, 5.589, 16.767 and 50.301 mg/L, respectively, when compared to the negative control. There was 21.83 % and 61.51 % inhibition of average growth rate and yield of alga, respectively, in the reference substance group at 0.9 mg/L. Observed cells were found morphologically normal.
