Clinical Observations and General Appearance:
Animals were observed for clinical signs, mortality, until sacrifice.
Onset, duration and severity of any sign were recorded. The clinical signs and mortality observations were conducted at 10, 30, 60 minutes, 2, 4 and 6 hours on the day of dosing and once daily thereafter for 14 day. Daily observation was done as far as possible at the same time.
The observations were included general clinical signs, observations of eyes, mucous membranes, respiratory, circulatory system and behavior pattern.
Body weights:
Individual animal body weights were recorded pre-test (prior to administration of the test item), day 7 and at termination on day 14.
Gross Pathology:
Necropsy was performed on animals surviving at the end of the study. Macroscopic examination of all the orifices, cavities and tissues were made and the findings were recorded. All animals surviving the study period were sacrificed by the carbon dioxide asphyxiation technique (day 15).

- Executive summary:
A study was designed and conducted to determine the dermal reaction profile of [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl] methylene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 84434-47-9) in Sprague Dawley rats. The study was performed as per OECD Guidelines 402 and complying to the GLP procedures.Ten rats (5 male and 5 female) were used for conducting dermal irritation/ corrosion study.
The animals were kept in their cages for at least 5 days prior to administration for acclimatization to the laboratory condition and after acclimatization period, animals were randomly selected. Approximately 24 hours before application, the hair of each rat was closely clipped from the trunk (dorsal surface and sides from scapular to pelvic area) with an electric clipper, so as to expose at least 10% of the body surface area. The test item was applied directly onto the exposed skin of the animal, taking care to spread the test item evenly over the entire area of approximately 10% of the total body surface area or as much of the area as can reasonably be covered. The test item was held in contact with the skin using a porous gauze dressing and non irritating tape around the animal to cover the exposure site for first 24 hours exposure period. Elizabethan collar was placed on each animal for first 24 hours after application of the test item. These collars prevent ingestion of test item. Following 24 hours of exposure, the wrapping was removed and the test site wiped free of excess test item. Distilled water was used to remove residual test item.
The test item [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl] methylene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 84434-47-9) was applied to shorn skin of 5 male and 5 female animals at 2000 mg/kg body weight. Administration of the test item at 2000 mg/kg did not result in any skin reaction at the site of application during the study period of 14 days. Also, the erythema and edema score of rats was calculated as 0. Administration of the test item did not result in any signs of toxicity and mortality during the study period of 14 days. Animals exhibited normal body weight gain through the study period of 14 days. Gross pathological examination did not reveal any abnormalities attributable to the treatment.
Hence, it was concluded that [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl]methylene] cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 84434-47-9) was Non-Irritating to the skin of Sprague Dawley rats under the experimental conditions tested and Classified as “Category- Not Classified” as per CLP Classification.

Skin Irritation:
In different studies, N-(4-{[4-(dimethylamino)phenyl][4-(methylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium acetate has been investigated for potential for dermal irritation to a greater or lesser extent. The studies are based on in vivo experiments in rabbits for target chemical and its closely related read across substances, terephthalic acid [CAS: 100-21-0], Isophthtalic acid [CAS: 121-91-5] and N-(4-{bis[4-(dimethylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium chloride [CAS: 548-62 -9].
A study was designed and conducted (Sustainability Support Services (Europe) AB, 2017) to determine the dermal reaction profile of [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl] methylene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 84434-47-9) in Sprague Dawley rats. The study was performed as per OECD Guidelines 402 and complying to the GLP procedures. Ten rats (5 male and 5 female) were used for conducting dermal irritation/ corrosion study.
The animals were kept in their cages for at least 5 days prior to administration for acclimatization to the laboratory condition and after acclimatization period, animals were randomly selected. Approximately 24 hours before application, the hair of each rat was closely clipped from the trunk (dorsal surface and sides from scapular to pelvic area) with an electric clipper, so as to expose at least 10% of the body surface area. The test item was applied directly onto the exposed skin of the animal, taking care to spread the test item evenly over the entire area of approximately 10% of the total body surface area or as much of the area as can reasonably be covered. The test item was held in contact with the skin using a porous gauze dressing and non irritating tape around the animal to cover the exposure site for first 24 hours exposure period. Elizabethan collar was placed on each animal for first 24 hours after application of the test item. These collars prevent ingestion of test item. Following 24 hours of exposure, the wrapping was removed and the test site wiped free of excess test item. Distilled water was used to remove residual test item.
The test item [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl] methylene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 84434-47-9) was applied to shorn skin of 5 male and 5 female animals at 2000 mg/kg body weight. Administration of the test item at 2000 mg/kg did not result in any skin reaction at the site of application during the study period of 14 days. Also, the erythema and edema score of rats was calculated as 0. Administration of the test item did not result in any signs of toxicity and mortality during the study period of 14 days. Animals exhibited normal body weight gain through the study period of 14 days. Gross pathological examination did not reveal any abnormalities attributable to the treatment.
Hence, it was concluded that [4-[[4-anilino-1-naphthyl][4-(dimethylamino)phenyl]methylene] cyclohexa-2,5-dien-1-ylidene]dimethylammonium acetate (CAS No. 84434-47-9) was Non-Irritating to the skin of Sprague Dawley rats under the experimental conditions tested and Classified as “Category- Not Classified” as per CLP Classification.
This result is supported by the experimentalthe study summarized inHazardous Substance Databank (HSDB), U S National Library of Medicine, last updated 2012; for theclosely related read across substance, terephthalic acid [CAS: 100-21-0].10 mL oily paste containing 80% terephthalic acid was applied to the equal sites on the hands of human volunteers (number not specified).The volunteers were observed for signs of irritation (duration not specified).
Terephthalic acid when applied as an oily paste was not irritating to human skin.
The above results are further supported by the experimental study summarized inHazardous Substance Databank (HSDB), U S National Library of Medicine, last updated 2012; for theclosely related read across substance, Isophthtalic acid [CAS: 121-91-5].500 mg undiluted isophthalic acidwas applied for 24 hr to abraded or unabraded skin of 6 albino rabbits that was previously clipped. The treated site was occluded. The rabbits were observed for signs of irritation and reactions were scored at 24 hours and 72 hours according to Draize method.With intact skin, mild erythema was noted in 1/6 animals at 24 hr (score=0.2/8), and 0/6 animals at 72 hr (score=0/8). With abraded skin, mild erythema was noted in 3/6 animals at 24 hr (score=0.5/8) and 0/6 animals at 72 hr (score=0/8). No edema was observed.
Based on these observations and scores, isophthalic acid was considered to be not irritating to albino rabbit skin.
These results are also supported by the experimental study summarized in Scientific Committee on Cosmetology (seventh series), 1988; for the closely related read across substance, N-(4-{bis[4-(dimethylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium chloride [CAS: 548-62-9]. 500mg of undiluted N-(4-{bis[4-(dimethylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium chloride was applied to rabbit skin and later observed for skin reactions (duration not specified). Since, N-(4-{bis[4 -(dimethylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium chloride failed to induce any skin reaction, it was considered to be not irritating to the rabbit’s skin.
Available data for the target and read across substances, indicate a strong possibility of N-(4-{[4-(dimethylamino)phenyl][4-(methylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium acetate being not irritating to skin.Comparing the above annotations with the criteria of CLP regulation,N-(4-{[4-(dimethylamino)phenyl][4-(methylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium acetatecan be classified under the category “Not Classified”.

In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the ocular irritation potential was estimated forN-(4-{[4-(dimethylamino)phenyl][4 -(methylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium acetate. N-(4-{[4-(dimethylamino)phenyl][4-(methylamino)phenyl]methylene}cyclohexa-2,5-dien-1 -ylidene)-N-methylmethanaminium acetate was not irritating to new Zealand White rabbit eyes.
This result is supported by the experimental study summarized inHazardous Substance Databank (HSDB), U S National Library of Medicine, last updated 2002; for theclosely related read across substance,t –butyl perbenzoate[CAS: 614-45-9]. 2 drops oft- butyl perbenzoatesolution in 50% dimethyl phthtalatewas instilled into rabbit eyes (number not specified).The rabbits were observed for signs of irritation (duration not specified) and graded on a scale of 0-7. The above studies are also supported by the experimental study summarized in EPA Publication 744-R-15-001; August 2015, for the closely related read across substance 6H-dibenzo[c,e][1,2]oxaphosphinine 6-oxide (DOPO) [CAS: 35948-25-5]. Neat test material (0.1 mL) was instilled in left eye of 3 female albino rabbits. Eyes were monitored for up to 7 days. Moderate signs of eye irritation that cleared in 7 days were observed among the rabbits. Since the effects were reversible by 7 days, 6H-dibenzo[c,e][1,2]oxaphosphinine 6-oxide (DOPO) can be considered not irritating to rabbit eyes.
Based on the available data for the target and read across substances and applying the weight of evidence approach, N-(4-{[4-(dimethylamino)phenyl][4-(methylamino)phenyl]methylene}cyclohexa-2,5-dien-1-ylidene)-N-methylmethanaminium acetate can be considered to be not irritating to eyes.Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Not Classified”.
