- Details on test material:
- A bulk sample (stated weight 300 g) of the test material was supplied by the Study Sponsor, arriving at Pharmaco LSR 26 July 1994. The test material was cream in colour and granular in form and was identified as M-3221, batch number 944002. The sample was stored at ambient temperature and protected from light until use in testing.
The identity, strength and purity of the test material received and its stability under the storage conditions were the responsibility of the Sponsor.
Nominal exposure concentrations quoted in this report refer to the test material as received; no allowance has been made for a purity of less than 100%.

- Details on test organisms:
- Axenic, uni-algal slope cultures of Selenastrum capricornutum, CCAP number 278/4 were obtained from the Culture Collection of Algae and Protozoa (CCAP), Freshwater Biological Association, Ferry House, Ambleside, Cumbria, 2 November 1994. These slopes were stored in an illuminated incubator nominally at 5°C until required.
Initially, liquid cultures for the test inoculum were established by inoculation of OECD mineral salts medium (50 ml; Appendix 1) with cells aseptically removed from slope cultures. Subsequently, subcultures were prepared from these liquid cultures by the aseptic transfer of aliquots into fresh OECD medium, which was then incubated for several days, until the required cell density had been achieved.

- Cell counts in control and test cultures in the rangefinding test indicated that the 72-hour EC5O of M-3221 to Selenastrum capricornutum was greater than 100 mg/l (nominal value).
The results of chemical analysis indicated that M-3221 was poorly soluble in the mineral salts medium; analysis of samples of filtered media was not attempted because a method for extracting M-3221 from water at low levels (< 1 mg/l) could not be identified. Based on visual observations, the limit of aqueous solubility was less than 1 mg/l. Analysis of unfiltered samples of test media indicated that at 1 and 10 mg/l the levels achieved were adequately maintained; measured concentrations at the other levels were variable.
Chemical analysis
The results indicate that the exposure concentration, although lower than intended (52% of its nominal value), was adequately maintained during the test; the overall mean measured level of M-3221 was 5.68 mg/l.
Algal growth
Exposure of Selenast rum capricornutum to M-3221 at a mean measured concentration of 5.68 mg/l did not result in a significant reduction (p >0.05) in either the average specific growth rates or biomass of test cultures compared to solvent control cultures.
Thus, the no-observed-effect concentration (NOEC) for growth rate and biomass was not identified but must be >5.68 mg/l and the median effect concentrations (ErC5O and EbC5O) could not be calculated.
