Justification for type of information:
n the absence of an algal inhibition study for the target substance, and in order to address the endpoint
requirement for Annex VII ECHA dossier submission for yogurtene, it is proposed to use (Q)SAR results in a weight of evidence approach.
Two well-established software tools have been used, ECOSAR and OECD QSAR Toolbox.
ECOSAR is a collection of QSAR models estimating aquatic toxicity, including short-term and long-term
toxicity to Algae. The ECOSAR models and underlying methodology have been developed over a period
of more than 25 years by EPA/OPPT (EPA, 2012). Most of the models are based on the relationship
between log Kow and toxicity (LC50 or EC50) and take into account different structural classes.
The OECD QSAR toolbox incorporates information and tools from various sources and is intended to be
used in filling gaps in (eco)toxicity data needed for assessing the hazards of chemicals. Acute aquatic
toxicity effects are the most data-rich endpoints and strategies for grouping chemicals to fill data gaps
to assess acute aquatic toxicity endpoints have been established (OECD, 2013).
