The dermal irritation potential of target chemical 2-Methoxybenzyl alcohol (CAS no: 612-16-8) was assessedin various experimental studies which were conducted on rabbits, guinea and humans for target chemical2-Methoxybenzyl alcohol (CAS no: 612-16-8) and its structurally similar read across substancesAnisic alcohol (CAS No: 105-13-5) and Methyl benzoate (CAS No: 93-58-3). The studies are based on in vivo experiments in rabbits, guinea and humans for target chemical2-Methoxybenzyl alcohol (CAS no: 612-16-8) and its structurally similar read across substancesAnisic alcohol (CAS No: 105-13-5) and Methyl benzoate (CAS No: 93-58-3).The predicted data using the OECD QSAR toolbox andDanish QSAR databasehas also been compared with the experimental data and summarized as below;
The D. Belsito et al., {Food and Chemical Toxicology, Vol. 50, Pg. no. S52-S99, 2012} conducted skin irritation studyto assess the skin irritation potential of target chemical 2-Methoxybenzyl alcohol (CAS no: 612-16-8) on human volunteers as a part of induction of human repeated insult patch test.The chemical was applied to the skin of each subjects at a dose of 1.2% in ethyl alcohol and later observed for skin reaction.Slight erythema was observed at one reading only in 4 out of 39 subjects. According to Danish QSAR database, skin irritation effects were estimated by using four different models i.e, Battery, Leadscope, SciQSAR and CASE Ultra for 2-Methoxybenzyl alcohol (CAS no: 612-16-8). Based on estimation, no severe skin irritation effects were known when 2-Methoxybenzyl alcohol (CAS no: 612-16-8) was exposed to rabbit skin. Hence, 2-Methoxybenzyl alcohol (CAS no: 612-16-8) can be considered to be not irritating to skin.
In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the skin irritation potential was estimated for test chemical2-Methoxybenzyl alcohol (CAS no: 612-16-8) .The chemical 2-Methoxybenzyl alcohol (CAS no: 612-16-8) is estimated to be not irritating to skin of albino rabbits.
TheD.W. SHARP {Toxicology, 9 (1978) 261-271} carried out thePreliminary irritation test study of read across substanceAnisic alcohol (CAS No: 105-13-5)in guinea pigs to determine the concentration suitableforthe sensitization study[injection challenge concentration (ICC) and application challenge concentration(ACC)].In the preliminary test, the guinea pigs were treated intradermally and dermally. 4 guinea pigs were (same sex) were injected intradermally in the clipped and shaved flanks with 0.1ml aliquots of a range of concentration of test substance in physiological saline. 24 hours later, the skin was examined for erythema and edema.The concentration giving slight but perceptible irritation with no oedema was selected as the injection challenge concentration (ICC).Aliquots of 0.1ml using a range of concentration of the test substance in distilled water were applied in small circular areas to the shaved flanks of 4 previously untreated guinea pigs.24 hours later, the skin was examined for erythema and the highest concentration which caused no irritation was selected as the application challenge concentration (ACC).As a result of preliminary studies, the concentrations of Anisic alcohol at which no irritation was observed were 0.25% for the ICC and 10% for ACC.Since the test chemical did not induce any cutaneous effects at these concentrations, the Anisic alcohol (CAS No: 105-13-5) was considered to be not irritating to the skin of guinea pigs.
The above results were further supported by the primary skin study conducted by Toshio Takenaka {Journal of J.C.C.A. Special Issue (1968) Pg 313–329} for read across substance Methyl benzoate (CAS No: 93-58-3) by mean of closed patch to examine the degree of skin irritancy caused by the chemical on 96 human subject. Methyl benzoate (CAS No: 93-58-3)was applied to the back, forearm and upper arm of 96 male/female humans in Japan under occlusive condition at a concentration of 0.05-0.5% 24–48 h and the sites evaluated 30 min after the patches were removed. Cream base or ethyl alcohol was used as a vehicle. None of the subjects showed any signs of skin irritation. ThereforeMethyl benzoate (CAS No: 93-58-3) was considered to be non-irritant to the skin.

TheBecker LC, et al., {International Journal of Toxicology,November/December 2012 vol. 31 no. 6 suppl 342S-372S} assessed an ocular irritation potential of read across chemicalMethyl benzoate (CAS No: 93-58-3)by performing rabbit eye test.Neat (100%) methyl benzoate was applied to the center of the cornea while the lids were retracted. One minute later the lids were released. The eyes were examined 18–24 h later in strong diffuse daylight and then stained with fluorescein.Methyl benzoate was given a grade 1 (iritis and slight internal congestion) after 24 hours of observation. Based on this rating, methyl benzoate can be considered as an eye irritant.
The above results were further supported by the experimental study summarized by Cosmetic Ingredient Review Expert Panel {Journal of the American College of Toxicology, Volume: 9 issue: 2, page(s): 259-277, March 1, 1990}of read across chemicalPhenoxyethanol(CAS no: 122-99-6) on three rabbits.The undiluted chemical was applied to theconjunctival sacs of the eyes of three rabbit. One eye of each rabbit was rinsedand later observed for ocular lesions.The unrinsed eyes had moderate to severe erythema and edema, injected irides, and slight corneal opacrties. The three unrinsed eyes also had fluorescein staining of the cornea and adnexa. The rinsed eyes had slight (two eyes) to moderate (one eye) irritation. The rinsed eyes had fluorescein staining of the adnexa.Since the chemical produced severe eye damage, Phenoxyethanol(CAS no: 122-99-6) was considered to be irritating to the eye of three rabbits.
Based on the available data for the target and read across substances and applying the weight of evidence approach, it can be concluded that chemical 2-Methoxybenzyl alcohol (CAS no: 612-16-8) is able to cause eye irritation and considered as irritating. Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Category 2”.
Justification for classification or non-classification
The skin and eye irritation potential of test chemical 2-Methoxybenzyl alcohol (CAS no: 612-16-8) and its structurally similar read across substanceswere observed in various studies. The results obtained from these studies indicate that the chemical 2-Methoxybenzyl alcohol is unlikely to cause skin irritation but can cause serious eye damage. Hence 2-Methoxybenzyl alcohol (CAS no: 612-16-8) can be classified under the category “Not Classified” for skin and “Category 2” for eye as per CLP.
