Skin irritation:
Various studieshas been investigated for the test chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)to observe the potential for dermal irritation to a greater or lesser extent. The studies are based on in vivo experiments in rabbits and guinea pigs for target chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) and its structurally similar read across substancesSodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6)andAcid red 180(CAS No:6408-26-0).The predicted data using the OECD QSAR toolbox has also been compared with the experimental data and summarized as below;
In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the skin irritation potential was estimated for test chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) .The chemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) is estimated to be not irritating to skin of New Zealand White rabbits.
The above result was further supported by the experimental skin irritation studies reported by European Commission (EC) - Scientific Committee on Cosmetology (SCC) {Environment and Quality of Life - Reports (Seventh Series), European Commission (EC) - Scientific Committee on Cosmetology (SCC), 1988} for two read across chemicals Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6) andAcid red 180(CAS No:6408-26-0) as follows;
The skin irritation study of chemicalSodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6) was conducted on intact and abraded skin of rabbits to assess the degree of skin irritation caused by the chemical. When rabbits were exposed with 100 mg of undilutedSodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6), no skin irritating effects were observed. Thus the chemicalSodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6) was considered to be not irritating to the rabbits’skin.
The next skin irritation studywas conducted on skin of guinea pigs via repeated applicationforchemicalAcid red 180(CAS No:6408-26-0). The repeated application of 1% solution ofAcid red 180 in 1% methylcellulose to the skin of each guinea pig (3 times on day 1, and 3 times on day 2) did not induce any skin reaction.Therefore the chemicalAcid red 180(CAS No:6408-26-0) was considered to be not irritating to the skin of guinea pigs.
Thus based on the available data for the target and read across substances and applying the weight of evidence approach, it can be concluded that chemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) is unable to cause skin irritation and considered as not irritating. Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Not Classified”.
Eye irritation:
In different studies,the test chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) has been investigated for potential for ocular irritationto a greater or lesser extent. The studies are based on in vivo experiments in rabbits for target chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) andits structurally similar read across substancessubstanceAcid Yellow 23 (CAS No: 1934-21-0)and Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6).The predicted data using the OECD QSAR toolbox has also been compared with the experimental data and summarized as below;
In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the eye irritation potential was estimated for test chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7).The chemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) is estimated to be not irritating to eye of New Zealand White rabbits.
The European Commission (EC) - Scientific Committee on Cosmetic Products and Non-Food Products (SCCNFP) {Opinion concerning: Acid Yellow 23 (Colipa n° C29); European Commission (EC) - Scientific Committee on Cosmetic Products and Non-Food Products (SCCNFP) SCCNFP/0786/04; 23 April 2004} reported anocular irritation study of read across chemical Acid Yellow 23 (CAS No: 1934-21-0) on New Zealand White rabbits. In this test,about 30 μl of test solution (3 % (w/v) Acid Yellow 23 in aqueous solution with 0.5 % (w/v) hydroxypropyl methylcellulose and 0.25 % (w/v) laureth-10 acetate) or vehicle were administered to the right eye of 6 female and 6 male rabbits, followed by closing the eye for approx. 1 sec to prevent substance loss. Ocular irritation was determined according to a modification of the Draize test. All animals were checked for viability twice daily. Ocular irritation was scored 24 h after each treatment and prior to the next instillation. On days 1, 3, 7, 14, and 21 the eyes were also evaluated 1 h after treatment. 24 h after each treatment additionally eye stain and particle depositions were scored. Ophthalmic observations were conducted on days 3, 7 and 14 always prior to instillation. The left eyes served as untreated controls.No lethality or significant clinical signs, and no substance-related weight change were observed. Except slight conjunctival redness or discharge, that were seen sporadically in the eyes of the animals, all animals were free of significant signs of ocular irritation. No significant signs of eye staining or particle depositions were observed. At ophthalmoscopic examinations, no ocular abnormities were noticed.Thus on the basis of observed ocular effects, the chemical Acid Yellow 23 (CAS No: 1934-21-0) was considered as not irritating to the New Zealand White rabbits’ eyes.
The above results were further supported by the experimental study reported by European Commission (EC) - Scientific Committee on Cosmetology (SCC) {Environment and Quality of Life - Reports (Seventh Series), European Commission (EC) - Scientific Committee on Cosmetology (SCC), 1988} for read across chemical Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6) on rabbits. When undilutedSodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonatewas installed into the eye of each rabbits, none of the treated rabbits produced any ocular lesions. Thus the chemicalSodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6) was considered to be not irritating to the rabbits’eye.
Thus on the basis of available data for thetarget chemicalDisodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)and its structurally similar read across substancesAcid Yellow 23 (CAS No: 1934-21-0)and Sodium 4-(3-hydroxy-5-methyl-4-phenylazopyrazol-2-yl)benzenesulphonate(CAS No:6359-82-6),it can be concluded thatchemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7) is unable to cause eye irritation and considered as not irritating. Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Not Classified”.
Justification for classification or non-classification
The skin and eye irritation potential of test chemical Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)and its structurally similar read across substanceswere observed in various studies. The results obtained from these studies indicate that the chemical 4-aminobenzene-1,2-dicarbonitrile is unlikely to cause skin and eye irritation. Hence Disodium [5-chloro-3-[[4,5-dihydro-3-methyl-5-oxo-1-(3-sulphophenyl)-1H-pyrazol-4-yl]azo]-2-hydroxybenzene-1-sulphonato(4-)]hydroxychromate(2-) (CAS No: 6408-31-7)can be classified under the category “Not Classified” for skin and eye as per CLP.
