The culture of D. subspicatus was obtained in October 2006 from the "Collection of alga", Institut für Pflanzenphysiologie of Universität Göttingen. The alga are kepts as stock culture on solid agar at 8°C. From an aliquot of this stock culture, the pre-culture was prepared.

In the main study, three concentrations of the test item ina geometric series (spaced by factor 10, the highest being 100 mg/L) were used. The treatments were used to incubate the unicellular freshwater green alga desmodesmus subspicatus for a period of 72 hours.
The cell concentration of each replicate was determined by measuring the absorption of the cuvettes at 440 nm every 24 hours with a spectral photometer. With these measured values, the number of cells was calculated (linear correlation between cell concentration and absorption given). Determination of 72h-EC50 of Montanov L against Desmodesmus subspicatus according to OECD 201 resp. EU C.3.
This study was performed in order to evaluate the toxic potential of Montanov L towards Desmodesmus subspicatus CHODAT, an unicellular freshwater alga. The incubation period of 72h corresponds to a subchronic test, as during this period, about five cell divisions take place. Selection of the test system was made following the proposal of the guidelines.
The study was conduced in accordance with the following guidelines:
OECD guideline for the testing of chemicals n° 201 "alga growth inhibition test"
EU-guideline C.3 "alga inhibition test"
As the test item is poorly soluble in water, the water accomodated fraction was tested. This was done by shaking the nominal load with the appropriate amount of deionised water for 24 hours, followed by membrane filtration.

The EC50 of potassium dichromate was tested in a curent reference test. The values lay within the normal range of the laboratory.
