- The test substance was made into a paste using Tween 80 as solubilizing agent. Water for dilution was added and the mixture homogenized using an Ultra-Turrax and ultrasonic bath and poured into the chamber under stirring with a glass rod. The chambers contents were then stirred for approximately 2 hours with a KPG-stirrer and a glass rod. The 220 mg/l test batch was present as clear solution. The 500 mg/l test batch showed a slight tubidity. Additionally substance deposits on the surface and on the buttom of the chamber were observed. In order to dtermine the substance concentration of the 500 mg/l group, water samples were taken from the middle of the test chamber from the first and third preparation of the test after 0 and 24 hours using a screw-neck, glass bottle. The analytical results were in a range of about 27,9 - 33,5 % of the nominal concentration.
- Details on test conditions:
- The studdy was conducted in a semistatic system. The fish were transferred each 24hours to new test chambers, which were calibrated to 10 liters, were made out of glass (length 30 cm, width 22 cm, height 24 cm) and stood in a water bath made out of HOSTALIT Z (R) with a Plexiglass viewing panel. The temperature of the water bath was regulated by a thermostat to 22 *_1 °C. The chambers were illuminated from above from 6.00 a.m. to 6.00 p.m.. The light intensity directly over the chambers was approximately 700 lux. The test chambers were not aerated during the course of the study.
