Description of key information

The toxicity of 5’-O-(4,4’-Dimethoxytrityl)thymidine was tested according to OECD guidelines 201 and 202 for acute toxicity studies with aquatic invertebrates (Daphnia magna) and algae (Pseudokirchneriella subcapitata). No toxic effect on D. magna was observed up to the limit of water solubility (NOEC ≥ 100 mg/L nominal, corresponding to 2.99 mg/L measured). For P. subcapitata a 72 h ErC10 of 2.975 mg/L (95 % C.L. 2.256 – 3.924 mg/L) was determined. The 72 h NOErC is 1.30 mg/L. An ErC50 could not be determined, as up to the limit of water solubility only about 10 % effect could be observed.

Toxicity to invertebrates:

Daphnia magna Straus was exposed for 48 hours to 5’-O-(4,4’-Dimethoxytrityl)thymidine in a static short-term toxicity study according to the OECD guideline 202 (2004) and in compliance with the OECD-GLP principles. The study was conducted as limit test with a measured concentration of 2.99 mg/L (limit of water solubility; nominal concentration of 100 mg/L). The test concentrations were determined analytically by HPLC-UV. As the measured test item concentrations are not within ± 20 % of the nominal loading rate, according to OECD 202 (2004) and OECD 23 (2000), all results are given in relation to the analytically measured test item concentrations.

No effect on D. magna was observed.

 

Toxicity to algae:

The freshwater algae Pseudokirchneriella subcapitata was exposed to 5’-O-(4,4’-Dimethoxytrityl)thymidine for 72 hours in a static toxicity study according to the OECD guideline 201 (2006) and in compliance with the OECD-GLP principles. In this study the72 h ErC20 (= 5.028 mg/L) is calculated by extrapolation slightly exceeding the solubility of the substance. The 72 h ErC50 (> 4.66 mg/L) could not be determined, as up to the limit of water solubility only about 10 % effect were observed.

The ErC10 value is interpolated and does not exceed the water solubility of the substance.

The test concentrations were determined analytically by HPLC-UV. 

The chemical analysis was conducted using the method with the lowest LOQ possible with the available equipment at the test site. The signal to noise ratio would be too low for a lower LOQ (in the range of or below the Limit of detection).
