Description of key information
Read-across data for the dipeptide L-alanyl-L-tyrosine is provided and used to derive an effect value for the UVCB substance reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride.
The reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride is not classified as skin sensitizer.
Key value for chemical safety assessment
Skin sensitisation
Endpoint conclusion
- Endpoint conclusion:
- no adverse effect observed (not sensitising)
- Additional information:
No data is available on the skin sensitization potential of the reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride.
The substance consists mainly of the dipeptide L-alanyl-L-tyrosine hydrochloride (38-42%) and potassium chloride (46-50%). Besides these main components only non-hazardous / non-classified amino acids/peptides in small amounts are present (e.g. L-alanine hydrochloride 4-7%). Based on the composition of the substance it can be assumed that the results obtained with the individual main components (L-alanyl-L-tyrosine and potassium chloride) apply likewise to the test material under investigation (reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride). Therefore read-across data for the dipeptide L-alanyl-L-tyrosine is provided and used to derive an effect value for the UVCB substance reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride. No data is available for the other components of the reaction mass. However, both KCl and L-Alanine are toxicologically well characterized and no skin sensitization potential is known.
L-Alanyl-L-Tyrosine (L-Ala-L-Tyr) is classified as inconclusive (based on the absence of a biologically relevant activation of the antioxidant/electrophile responsive element (ARE)-dependent pathway in keratinocytes at test concentrations < 1000 μM) in the KeratinoSens assay.
L-Alanyl-L-Tyrosine (L-Ala-L-Tyr) is classified as negative (no increase in the expression levels of CD86 cell surface marker in the U937 cell line) in the U-Sens™ assay.
Therefore, the reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride is not classified as skin sensitizer.
Respiratory sensitisation
Endpoint conclusion
- Endpoint conclusion:
- no study available
Justification for classification or non-classification
Read-across data for the dipeptide L-alanyl-L-tyrosine is provided and used to derive an effect value for the UVCB substance reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride.
L-Alanyl-L-Tyrosine (L-Ala-L-Tyr) is classified as inconclusive in the KeratinoSens assay and negative in the U-Sens™ assay. No data is available for the other components of the reaction mass. However, both KCl and L-Alanine are toxicologically well characterized and no skin sensitization potential is known.
Therefore, the reaction mass of L-alanyl-L-tyrosine hydrochloride and potassium chloride is not classified as skin sensitizer.
