EFFECT PARAMETERS MEASURED
Observations of the fish were made after 2, 24, 48, 72 and 96 h of exposure. Mortality of the fish was recorded at the same time intervals.
Dead fish were removed immediately from the test medium and weight and length recorded.
Fish were considered to be dead when they showed no reaction after touching the caudal fin or no visible breathing movements.

- During the test, all fish were dead at the nominal concentration of 100 mg/l after 48 h of exposure. No mortality was observed at 10 and 1 mg/l and in the control after 96 h of exposure.

Based on these data the nominal lethal concentration (LC50) of test item was estimated to be 10-100 mg/l.

The acute lethal toxicity of test item to Poecilia reticulata (Guppy) was investigated under static exposure conditions over a period of 96 h. The applied nominal test concentrations were 1, 10 and 100 mg/l. Therefore, the study represents a screening test. The individual test concentrations were prepared by dilution of a 200 mg/l stock solution. However, the test material was not completely dissolved. No chemical analyses of the test concentrations were conducted.
During the test, all fish were dead at the nominal concentration of 100 mg/l after 48 h of exposure. No mortality was observed at 10 and 1 mg/l and in the control after 96 h of exposure.

The acute lethal toxicity of Fluorescent Brightener 312 to Danio rerio and Poecilia reticulata was investigated in two different experiments, both conducted under static exposure conditions and over a period of 96 h.
10 Danio rerio fish were exposed to each one of the follwoing nominal dose levels: 10, 18, 32, 58 and 100 mg/l. The lower test concentration causing 100 % of mortality was 58 mg/l, while the highest test concentration causing no mortality was 18 mg/l. The LC50 was estimated to be 41 mg/l.
The applied nominal test concentrations in the case of Poecilia reticulata were 1, 10 and 100 mg/l. During the test, all fish were dead at the nominal concentration of 100 mg/l after 48 h of exposure. No mortality was observed at 10 and 1 mg/l and in the control after 96 h of exposure. Other toxic effects than mortality, e.g. loss of coordination, hypoactivity and swimming on the back, were not observed. Based on these data the nominal lethal concentration (LC50) of test item was estimated to be 10-100 mg/l.
In both the cases, no chemical analysis of the test substance concentrations was conducted. In the first mentioned study, concentration-dependent slight haze and bluish opalescent shimmer were observed in all assays; in the test concentrations 32 - 100 mg/l, a slight to strong foaming occurred and after 24 hours a slight concentration-dependent substance precipitate at the edge of the aquaria was found from the test concentration 32 mg/l.
The test material resulted to be not completely dissolved also in the case of the other experiment.
