Read-across was performed in order to assess the acute toxicity of Ammonium [trisulphamoyl-29H,31H-phthalocyaninesulphonato(3-)-N29,N30,N31,N32]cuprat e(1-) (CAS 94534-99-3) to fish. The substance belongs to the chemical category of Phthalocyanines whose category members resemble in their chemical structure as well as in their physico-chemical properties. Thus, read-across data from analogous substances of the phthalocyanine category were used to cover this endpoint.
The existing data on Direct Blue 199 sodium salt (Direct Blue 199_Na; CAS 90295-11-7), Direct Blue 199 (CAS 90295 -08 -2) an
d Heliogen Blue 6920 (CAS 147-14-8) have been taken into account. The test procedures are scientifically acceptable. However, many details on the majority of the studies are not reported. Therefore, the assessment by the available read-across data is based on a weight-of-evidence approach:
Direct Blue 199 (CAS 90295 -08 -2) was tested for short-term toxic to fish:
- In 1974, Huntsman Textile Effects (Germany) GmbH conducted a study using Oncorhynchus mykiss as test organism; detailed data on the methods are not documented for this study. Acute toxicity for trout was tested at test concentrations of 100, 200 and 1000 mg/l at 24 and 48 hours. The 48h LC50 was determined to be >1000 mg/L (nominal).
Direct Blue 199_Na (CAS 90295 -11-7) was tested for short-term toxic to fish at different lot concentrations in several studies:
- In 1977, Huntsman Textile Effects (Germany) GmbH conducted another study using Oncorhynchus mykiss as test organism; detailed data on the methods are also not documented for this study. Acute toxicity for trout was tested at test concentrations of 100, 200 and 1000 mg/l at 24 and 48 hours. The 48h LC50 was determined to be >1000 mg/L (nominal).
- In 1978, Huntsman Textile Effects (Germany) GmbH conducted a study similar to OECD TG 203; the acute fish toxicity testing was performed according to published methods, which were adapted and standardized for the specific needs of the company, well before Test Guidelines for these methods were established by the OECD beginning 1981. These methods were comparable to today’s standard testing procedure published by the OECD within Guideline for Testing of Chemicals 203. Oncorhynchus mykiss was used as test organism. Acute toxicity for trout was tested at test concentrations of 100, 200 and 1000 mg/l at 24 and 48 hours. The 48h LC50 was determined to be 450 mg/L (nominal).
- In 1989, Huntsman Textile Effects (Germany) GmbH conducted a study similar to OECD TG 203; the acute fish toxicity testing was performed according to published methods, which were adapted and standardized for the specific needs of the company, well before Test Guidelines for these methods were established by the OECD beginning 1981. These methods were comparable to today’s standard testing procedure published by the OECD within Guideline for Testing of Chemicals 203. Danio rerio was used as test organism. Acute toxicity for zebrafish was tested at test concentrations of 100, 300 and 1000 mg/l at 24 and 48 hours. The 48h LC50 was determined to be >1000 mg/L (nominal).
The acute toxicity of Heliogen Blue 6920 (CAS 147-14-8) to fish was tested in a limit test performed in a static system with Danio rerio according to OECD 203. The 96h LC50 was determined to be >100 mg/l (BASF AG, 2004).
Conclusion:
Summing up, the 48h LC50 values determined for the source substances Direct Blue 199 sodium salt (Direct Blue 199_Na; CAS 90295 -11 -7) and Direct Blue 199 (CAS 90295 -08 -2)
ranged from 450 to >1000 mg/L (nominal). The 96h LC50 determined for the source substance Heliogen Blue 6920 (CAS 147-14-8) was >100 mg/L. Based on the available read-across data, Ammonium [trisulphamoyl-29H,31H-phthalocyaninesulphonato(3-)-N29,N30,N31,N32]cuprat e(1-) (CAS 94534-99-3) is assessed to be with high probability acutely not harmful to fish.Information on Registered Substances comes from registration dossiers which have been assigned a registration number.
