Test solutions

Vehicle:
yes
Details on test solutions:
For test concentrations prepared in the rangefmding test, stock solutions were prepared at concentrations of 10, 50 and 100 mg/L by adding the required nominal amount of test substance in the diluent, moderately hard synthetic freshwater, and mixing the solution for approximately 1 hour. For test concentrations prepared in the defmitive test, stock solutions were prepared at concentrations of 10, 20, 40, 80 and 160 mg/L by adding the required nominal amount of test substance in the diluent, and mixing the solution for approximately 2 hours.

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
Daphnia magna, one of the daphnid species specified in the OECD test guideline was maintained in the STILLIVIEADOW, Inc. culture laboratory. The Daphnids were No greater than 24 hours old at dosing. The environmental conditions were 16 hours light/8 hours dark cycle with a dissolved oxygen concentration of 60-105% saturation at dosing and temperature of 21°C.

Study design

Test type:
static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
48 h
Post exposure observation period:
None

Test conditions

Hardness:
Total hardness of 235 mg/L CaCO3.
Test temperature:
21°C
pH:
7.9
Dissolved oxygen:
Dissolved oxygen concentration of 60-105% saturation
Salinity:
No data
Nominal and measured concentrations:
10, 20, 40, 80 and 160 mg/L nominal.
Details on test conditions:
Exposure was conducted in 250-mL glass beakers filled with 200 mL of the appropriate solution of moderately hard synthetic freshwater. Each test concentration consisted of three replicates of ten daphnids. Three replicates of ten daphnids were not exposed to test substance and served as controls to demonstrate the condition of the test population. At 24 and 48 hours following dosing, each beaker was examined for immobility, and the number of mobile dapbnids was recorded.

Reference substance (positive control):
no

Results and discussion

Effect concentrations
Duration:
48 h
Dose descriptor:
EC50
Effect conc.:
ca. 29.8 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mobility
Remarks on result:
other: 95% confidence Limits = 28.6-30.2 mg/L.
Details on results:
Mobility of the Daphnids was 97-100% at 10 and 20 mg/L of test substance after 48 hr of exposure. There was 0% mobility at the test substance concentrations of 40, 80 and 160 mg/L at 48 hr. Mobility of the Daphnids at 160 mg/L test substance was 16.7% after 24 hr of exposure.
Reported statistics and error estimates:
The NOEC (No Observed Effect Concentration) and EC50 (median effective concentration) were determined by a commercially available statistical program (ToxCalc Version 5 .OTM). The Probit model was selected for the determination in this case. The 48 hr EC50 95% Confidence Limits were 28.6-30.2 mg/L.

Validity criteria fulfilled:
yes
Conclusions:
The 48 hr EC50 for Daphnia magna was approximately 29.8 mg/L with 95% Confidence Limits of 28.6-30.2 mg/L. The 48 hr NOEC was 20 mg/L.
Executive summary:
