PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: A stock solution (and the highest test concentration) of PF-03394197 with a nominal concentration of 56 mg/L was prepared by the addition of a nominal 0.112 g of test substance to dilution water made up to 2000 ml. The stock solution was ultrasonicated for 30 minutes after which it was observed to be a clear and colourless solution.
Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: freshwater crustacean, Daphnia magna
- Age at study initiation (mean and range, SD): <24 hours old
- Source: continuous laboratory cultures held at Brixham Environmental Laboratory.

ACCLIMATION
- Acclimation conditions (same as test or not): same
- Type and amount of food: fed on a mixed algae diet of Chlorella vulgaris, strain CCAP 211/12 and Pseudokirchneriella subcapitata, strain CCAP 278/4.
- Feeding frequency: fed daily ad libitum depending on age and density of the culture.

Nominal and measured concentrations:
Nominal concentrations: 0.56, 1.0, 1.8, 3.2, 5.6, 10, 32 and 56 mg/L
Measured concentrations: 0.55, 0.99, 1.8, 3.2, 5.6, 10, 33 and 56 mg/L
Details on test conditions:
TEST SYSTEM
- Test vessel: Glass beakers of 250 ml nominal capacity

- Renewal rate of test solution (frequency/flow rate): no
- No. of organisms per vessel: 5
- No. of vessels per concentration (replicates): 4
- No. of vessels per control (replicates): 4
- No. of vessels per vehicle control (replicates):
- Biomass loading rate:

OTHER TEST CONDITIONS
- Adjustment of pH: no
- Photoperiod: 16 hours light: 8 hours dark, with 20 minute dusk and dawn transition periods
- Light intensity:

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) :
An assessment of the response of the D. magna was made 24 and 48 hours after the start of the test. Each D. magna was viewed by eye and was defined as affected if showing no whole body movement, relative to the water, within a period of 15 seconds even if movement of individual appendages was visible. D. magna so affected were termed immobile. Any overt symptoms of toxicity were also recorded.
Reference substance (positive control):
no
Key result
Duration:
48 h
Dose descriptor:
EC50
Effect conc.:
18 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mobility
Details on results:
Percentage immobilised at 48 h:
Nominal 32 mg/L: 100%;
Nominal 56 mg/L: 100%
Validity criteria fulfilled:
yes
Conclusions:
The 48 hours EC50 of the test item is determined as 18 mg/L.
Executive summary:

The acute toxicity of the test item to Daphnia magna was determined, according to OECD Guideline 202 under GLP. The 48 hours EC50 of the test item is determined as 18 mg/L.
