6.3.2 Animal Husbandry
Daphnia magna is bred in the LAUS GmbH throughout the year. The animals are kept for the use in toxicity tests. They multiply by parthenogenesis, thus being genetically identi-cal. The husbandry is performed similar to the method described in the OECD guideline, following SOP 115 002 01 („Zucht und Hälterung von Daphnia magna STRAUS“), version 12 from 02. Feb. 2015.

- The study was performed as a limit test using 1 concentration at 100 mg/L. For the test concentration, 20 daphnia were exposed to the test item for 48 hours in a static test sys-tem. After 24 and 48 hours, the immobilised daphnia were counted.
Only one daphnia was immobilised in the tested concentration which can be stated as not significant. None of the animals was immobilised in the blank control - Results with reference substance (positive control):

In a GLP compliant 48 h acute toxicity test, the test substance was assessed according to OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test); and EU Method C.2 (Acute Toxicity for Daphnia).Daphnia magna were exposed to the test substance at concentrations of 100 mg/l and observed at 24 and 48 h for mobility. The EC50 of the test item in Daphnia magna after 48 hours is determined to be superior to 100 mg/L.
The EC50 of the test substance to Daphnia magna is >100 mg/l. Hence, the Blendazo Red is not toxic to aquatic invertebrates Daphnia magna.

Only one daphnia was immobilised in the tested concentration which can be stated as not significant (below validity criterion of maximum 10% immobility in blank control). In the blank control, none of the daphnia were immobilised (see table below).

Additional information
The prupose of the study was to to evaluate the toxic potential of Blendazol Red Blendwell towards freshwater flea, using the species Daphnia magna Straus, which was chosen in the guideline as a typical part of zoo-plankton.
Young daphnia, aged less than 24 hours at the start of the test, were exposed to the test item at a range of concentrations for a period of 48 hours.Immobilisation was recorded after 24 hours and 48 hours and compared with control values. The results were analysed in order to calculate the EC50at 24h and 48h.
The EC50 24h and 48h was > 100 mg/L, accordingly the test item is not toxic to aquatic invertebrates
