Concentrations were determined at the beginning and at the end of the test period. During the range finding study and main study, analytical samples (plus retain samples) were taken from a randomly chosen replicate per treatment group at beginning and at the end of the test (i.e. at 0 hour and 7 days). About 10 ml of sample was collected from all test concentration in duplicates.

Stock solution was freshly prepared in the Swedish Standard (SIS) growth medium. Required quantity of test item was taken in a beaker, in which small volume of Swedish Standard (SIS) growth medium was added; the solution was stirred well using glass rod. After complete solubility, test formulation was transferred to measuring cylinder, beaker was rinsed with Swedish Standard (SIS) growth medium and transferred again to measuring cylinder; the rinsing procedure was repeated until the complete transfer of test contents. Finally, the volume made up to required volume using Swedish Standard (SIS) growth medium.
A homogeneous distribution of the test item within the stock solution was achieved by stirring on magnetic stirrer before preparation of the test medium.
The test concentrations were prepared by adding appropriate amount of stock solutions and made up to required volume using Swedish Standard (SIS) growth medium.

TEST ORGANISM
- Source: in house maintained culture of Lemna minor, obtained from the Department of Botany, Tumkur University. Lemna minor culture is periodically sub cultured and maintained with the illumination and temperature of 6500 to 10000 lux and 22 to 26°C respectively. Lemna minor was maintained separately on Steinberg medium and Swedish Standard (SIS) media.

ADAPTATION
- Pre-culturing period: the pre culture was initiated 7 days before start of the test.
- Culturing media and conditions: 10 beakers containing 100 ml of culture medium; pre cultures are incubated under same conditions as the test cultures.

TEST SYSTEM
- Test vessel: sterilized, glass beakers of 150 ml capacity.
- Fill volume: the volume of the test solution at the initiation of the test in the test vessel was 100 ml.
- Cover: the test vessels were covered, allowing necessary air exchange, to minimise accidental contamination.
- No. of colonies per vessel: the initial Lemna minor colonies comprising 3 fronds.
- No. of fronds per colony: the initial number of frond number in all treatment replicates on day 0 was 12.
- No. of vessels per concentration: three replicates.
- No. of vessels per control: six replicates.

EFFECT PARAMETERS MEASURED
- Counting of frond number: fronds number at the start on day 0, day 4 and on day 7 was counted in each test vessel and recorded.
- Observation of fronds: fronds number appearing normal or abnormal was determined at beginning of test and on day 3, day 6 and at end of the test. Changes in plant development were recorded. Changes in plant development (e.g. frond size, appearance, indication of necrosis, chlorosis or gibbosity, colony break-up or loss of buoyancy and in root length and appearance) was recorded.
- Measurement of dry weight: dry weight was determined at test start from representative sample of the inoculum culture. Consistent with the approach taken in the test. At the end of the test, the plant material from each replicate of test concentration and control vessel was used to determine dry weight. To determine the dry weight they are blotted to remove excess water and then dried at 60 °C to a constant weight. Root fragments were included and dry weight was measured.

ENVIRONMENTAL PARAMETERS MEASUREMENT
The temperature (culture medium) and light intensity (at points the same distance from the light source as the Lemna fronds) was monitored once in a day during pre-culture and exposure during range finding study and main study. pH values of test concentrations (fresh and spent) were measured on day 0 and on day 7 at the termination of the test during range finding and main study.

RANGE-FINDING STUDY
- Test concentrations: 0.01, 0.1, 1, 10, 50 and 100 mg dyestuff/l.
- No. of colonies per vessel: initial Lemna minor colonies comprising 3 fronds.
- No. of fronds per colony: the initial number of frond number in all treatment replicates on day 0 was 9.
- No. of vessels per concentration: two replicates.
- No. of vessels per control: four replicates.
- Test condition: the temperature was found to be 24.1 to 24.8 °C and light intensity of 6840 to 6919 lux during pre-culture and exposure; the pH of test concentrations was found to be 6.43 to 6.51.
- Results used to determine conditions for the definitive study: the percentage of inhibition in growth rate resulted to be 0.0, 4.4, 13.2, 24.0, 62.3 and 100 based on the frond number at the concentrations of 0.01, 0.1, 1, 10, 50 and 100 mg dyestuff/l, nominal; the percentage of inhibition in yield resulted to be 0.9, 10.6, 27.4, 44.2, 82.3 and 100 based on the frond number at the concentrations of 0.01, 0.1, 1, 10, 50 and 100 mg dyestuff/l, nominal.

VALIDITY CRITERIA OF THE TEST
For the test to be valid, the doubling time of frond number in the control must be less than 2.5 days (60 h), corresponding to approximately a seven-fold increase in seven days and an average specific growth rate of 0.275 d-1.

Fronds appeared normal until day 3 and some fronds appeared normal and chlorosis on day 6 and 7 at the nominal tested concentration of 50 mg dyestuff/l. Chlorosis was observed on day 3 and chlorosis and desiccation was observed on day 6 and 7 at the nominal tested concentration of 100 mg dyestuff/l.

Measurement of dry weight: on day 0, dry weight was found to be 0.0214 g; at the end, mean dry weight of each group was found to be 0.231 g in control and 0.227, 0.197, 0.167, 0.134, 0.054 and 0.019 g at the nominal tested concentration of 0.01, 0.1, 1, 10, 50 and 100 mg dyestuff/l.

MEASURED CONCENTRATIONS
During the range finding and main study, samples from test concentration were analyzed on day 0 and on day 7. aConcentrations resulted to be within the acceptable range ± 20 % recovery to the nominal concentration.
Results with reference substance (positive control):
The growth inhibition test with Lemna minor exposed to 3,5-Dichlorophenol -97 %-, over 7 days the ErC50 values for growth rate of frond number and biomass (dry weight) were calculated to be 3.66 mg/l and 3.26 mg/l, respectively.
The ErC50 value for the frond number of 3.66 is in good agreement with the results of an international ring test between 2.7 and 3.4 mg/l (n=52).
Hence, the test with toxic standard establishes the acceptability of test system response and test procedures followed.

During the main study, all fronds appeared normal on day 0, 3, 6 and 7 in control and at nominal test concentrations of 0.9, 2.0 and 4.2 mg dyestuff/l.
