Additional information

The test substance has a reasonable water solubility, a log POWand a low molecular weight. Furthermore, bioaccumulation and hydrolytic instability is not expected. The physico-chemical properties of the substance and consideration of Lipinsky’s rule of 5 impose that oral absorption can be expected. The absorbed substance is subject to hepatic metabolic degradation. The following main metabolic steps are suggested to occur: 

-         Glucuronidation of the primary as well as the secondary alcohol

-         Oxidation of the primary/secondary alcohol (ketone formation)

-         Oxidative N-dealkylation

Discussion on bioaccumulation potential result:

No experimental data are available. However, physico-chemical and repeat-dose toxicity data allow for a prediction of the toxicokinetic behaviour of the substance. The substance is expected to be well absorbed and undergo metabolic changes in the liver. It is suggested that one important metabolic degradation step of HE-HTMP is the oxidative N-dealkylation, leading to the HTMP structure, the substance serving as the read-across candidate.
