The test item had a significant inhibitory effect on the AUC of the algae after the test periods of 72 and 96 hours at the concentration of 2.2 mg/L and at all higher test concentrations (results of Williams and Welch t-tests, one-sided,α= 0.05). Thus, this concentration was determined to be the 72 and 96-hour LOEC.
The 72 and 96-hour NOEC could not be determined since at the lowest nominal test concentration of 2.2 mg/L the AUC of the algae was significantly lower than in the control. The 72- and 96-hour EC50 values for the inhibition of AUC, average growth rate and yield and their 95% confidence intervals were calculated by Probit Analysis.
The microscopic examination of the algal cells at the end of the test showed no difference between the algae growing at the nominal test concentration of 46 mg/L and the algal cells in the control. The shape and size of the algal cells was obviously not affected by the test item up to at least this concentration.
In the control the biomass increased by a factor of 121 over 72 hours. The validity criterion of increase of biomass by at least a factor of 16 within three days was fulfilled. The mean coefficient of variation of the daily growth rates in the control during 72 and 96 hours was 4.4 and 32%, respectively.
According to the OECD test guideline, the mean coefficient of variation must not be higher than 35%. Thus, the validity criterion was fulfilled. The coefficient of variation of the average specific growth rates in the replicates of the control after 72 and 96 hours was 0.7 and 0.5%, respectively. According to the OECD test guideline, the coefficient of variation must not be higher than 7%. Thus, the validity criterion was fulfilled.

The influence of the test substance on the growth of the freshwater green algal species Pseudokirchneriella subcapitata was investigated in a 96-hour static test under GLP and according to OECD Guideline 201 (2006), by Höger (2010). The nominal concentrations of the test item of 2.2, 4.6, 10, 22, 46 and 100 mg/L were tested in parallel with a control. No analytical measurements were performed in the test. The 96h ErC50 was found to be 63 mg/L (60-67 mg/L, CI=95%), and a NOEC(growth rate)=10 mg/L determined, based on nominal concentrations.

The nominal concentrations of the test item of 2.2, 4.6, 10, 22, 46 and 100 mg/L were tested in parallel with a control. The measured concentrations of the test substance in the test media for 2.2 to 100 mg/L were between 100 and 108% of the nominal values at the start of the test, and between 100 and 107% at the end of the test. Thus the correct dosing, and stability of the substance in the test media, was confirmed.The 96h ErC50 was found to be 69mg/L (66-72mg/L, CI=95%), and a NOEC(growth rate)=22 mg/L determined, based on nominal concentrations.
The two available algae studies are in excellent agreement for growth rate and yield at 96-hours. Some parameters in the study by Liedtke (2010) could not be determined at 72-hours, and the 96-hour AUC could not be determined.
The chemical structure is not expected to be hydrolytically unstable, nor is there experimental evidence to suggest this. Stability of salicylonitrile in the test media was confirmed by Liedtke with analytical monitoring, and considering the good agreement between the effect concentrations in the two studies, it can be concluded that the results determined by Höger are fully reliable, despite the lack of measured concentrations. Because the latter results are slightly lower, and more conservative, these are the values taken forward for risk assessment and classification.
