- Stock culture: algae stock cultures were started by inoculating growth medium with algal cells from a pure culture on agar. The suspensions were continuously aerated and exposed to light in a climate room at 23 +/- 2°C.
Pre-culture: 4 days before the start of the test, cells from the algal stock culture were inoculated in culture medium at a cell density of 20000 cells/ml. The pre-culture was maintained under the same conditions as used in the test. The cell density was measured immediately before use. - Vehicle:

- A stock solution of JAVANOL was prepared at 100 mg/mL in acetone. A 1 in 10'000-dilution was performed with ISO Medium and stirred for 24 hours followed by filtration through a 0.2 um filter.

The study procedures described in this report were based on the EEC directive 92/69, publication no L383 part C-3 adopted December 1992; OECD guideline No. 201, Adopted june 7, 1984; and ISO standard 8692, first edition, 15 November 1989.
Javanol is a clear viscous liquid which is hardly soluble in water. The water solubility of Javanol is 3.8 mg/l (at 20°C) according to data of the sponsor. The procedure for preparation was based on research by the Analytical departement during method development and validation. This procedure included filtration of the stock solution through a 0.2 micrometer filter. Before filtration the test medium was exposed for one day to the test substance under continuous stirring to ensure maximum saturation.
After a range-finding test, two full tests were performed. The test solutions contained various amounts of a 0.2 micrometer filtrate prepared at nominal 10 mg/l with acetone as presolvent (0.1 ml/l). Analytical support showed that the algal suspensions were exposed to a range of initial Javanol concentrations of 0.4 to 4 mg/l in the first test and of 0.02 to 2 mg/l in the second and final test. The initial cell density was 10^4 cells/ml. The total test period was 72 hours.
Analysis of the samples taken at the end of the first full test showed that the actual test concentrations could not be maintained at 80% of the initial concentration, including the concentration in the solution without algae (76%). Instability increased with decreasing concentration. However, the analytical results of the second study showed that actual concentrations remained at 80% of initial in samples taken from the highest concentration without algae and in reserve samples taken from the lower concentrations. Hence EC50 values could be based on average concentrations.
