Materials and methods
- Principles of method if other than guideline:
- For this test substance, the partition coefficient cannot be determined according to OPPTS 830.7550 or OPPTS 830.7570 and a specific test plan was performed (see Section 7, Amendment #1). In this case, a log Pow was calculated based on experimental data obtained from the solubility of the test substance in water and n-Octanol. The solubility of the test substance in water and n-Octanol was determined separately by visual assessment according to the preliminary test from OPPTS 7840, using the calculation below.
Pow = [n-octanol (g/L)]/water (g/L).
n-Octanol solubility determination:
Preliminary Procedure: A preliminary assessment of the solubility of the test substance in octanol was conducted. Approximately 0.1 grams of the test substance was accurately weighed into a glass stoppered graduated cylinder for each solvent tested. Increasing volumes of octanol were added to the graduated cylinders containing the test substance. After each addition to the indicated volume, the graduated cylinder was vigorously shaken for 10 minutes. Visual observations were made for complete dissolution of the test substance. The solvent solubility of the test substance in the solution was demonstrated to be 50-100 g/L for water and <0.33 g/L for octanol, respectively. Duplicate test substance samples were prepared by weighing ~0.01 grams into separate
50 mL graduated cylinders and incremental measured volumes of octanol were added until complete dissolution was achieved. After each addition of octanol, the mixture was shaken vigorously and then stored at the test temperature (20°C) with intermittent
shaking for at least 30 minutes.
