The available data on both Direct Blue 251 and the structural analogues Similar Substance 01 and Similar Substance 02 have been considered for the aquatic toxicity assessment and classification.
The usage of information on the analogues can be considered as suitable and appropriated because the structural differences are expected to not influence the ecotoxicological properties; details on the approach followed are included in the document attached to the IUCLID section 13.
Direct Blue 251 is not rapidly degradable; however, on the basis of the chemical structure characteristics, the substance is expected to be possess a low potential for bioaccumulation.
Data are available on Direct Blue 251 for short-term toxicity to fish (Brachydanio rerio); data regarding both short and long-term toxicity to Daphnia magna and toxicity to Pseudokirchneriella subcapitata are available on the analogues.
Similar Substances 02 resulted to be not harful/toxic for algae and the same behavoiur is expected for Direct Blue 251.
Regarding short-term toxicity to Daphnia magna, Similar Substance 01 did not show acute toxic effects up to solubility limit in test water (i.e. 2.1 mg/l). Comparable results, in terms of solubility and effects, were obtained into the experiment perfromed on the Similar Substance 02. In addition, a long-term toxicity test is also availavle, in which No Observed Effect Concentration (21d) resulted to be higher than 10 mg/l (nominal), based on mortality and reproduction.
All experiments results have been considered in order to assess the aquatic toxicity potential of the substance.
In all cases, L(E)C50 values recorded resulted to be higher than 1 mg/l, thus the substance can be considered as not meeting the criteria to be classified for acute aquatic hazard.
Adequate chronic toxicity data are available on Daphnia magna, which indicate the NOEC as higher than 1 mg/l, thus the substance can be considered as not meeting the criteria to be classified for long-term aquatic hazard.
