Several methods are available for the determination of the partition coefficient n-octanol/water (Pow) of a substance: the shake flask method, the HPLC method, the estimation method (based on the ratio of octanol and water solubilities) and the calculation method (based on a QSAR). The shake flask method and the HPLC method are not suitable for surface active substances. For this type of substances, the calculation method or the estimation method are recommended.
Although the registered substance proved to be surface active (cf. 4.10 Surface tension), the calculation method was not applicable as the substance is a salt. The estimation methods was not applicable as well as it was not possible to precisely determine the solubility of the substance in both water and octanol. Indeed, the substance showed to be very miscible in water and to forms a viscous gel in octanol without saturated phase.
As the recommended methods were not applicable, the partition coefficient of the registered substance was determined by the shake flask method in a study in line with the standardised OECD Guideline 107. An analytical method was developed and validated to quantify the test item in water and in octanol phases. The results obtained in this study are in accordance with the validity criteria of the OECD Guideline 107. Six partition coefficient values were obtained (three duplicate runs with different solvent ratios); these values felt within a range of ± 0.3 units.
